TMIGD1通过促进细胞黏附抑制胃癌细胞侵袭、迁移能力的研究

Study on TMIGD1 inhibiting the invasion and migration of gastric cancer cells by promoting cell adhesion

目的探究免疫球蛋白结构域的跨膜蛋白1(TMIGD1)在胃癌组织中的表达情况及其与胃癌细胞侵袭、迁移、黏附的关系。方法收集60份胃癌患者病理组织标本,通过免疫组化染色检测TMIGD1的表达情况,并分析其与患者临床指标的关系。进一步挖掘TCGA数据库,通过Transwell、Western Blotting、细胞黏附率实验探究过表达TMIGD1对胃癌细胞侵袭、迁移、黏附的影响。结果TMIGD1在胃癌组织中呈低表达,TMIGD1阳性组黏液腺癌、发生淋巴脉管浸润的占比低于TMIGD1阴性组(P<0.05)。过表达TMIGD1可明显抑制胃癌细胞株AGS与BGC803的侵袭、迁移能力,提高细胞黏附能力(P<0.05)。结论TMIGD1在胃癌组织中呈低表达,过表达TMIGD1能够通过促进胃癌细胞黏附来抑制细胞侵袭、迁移。

Objective To explore the expression of transmembrane and immunoglobulin domain containing 1(TMIGD1)in gastric cancer tissues and its relationship with the invasion,migration and adhesion of gastric cancer cells.Methods A total of 60 pathological tissue samples from patients with gastric cancer were collected,the expression of TMIGD1 was detected by immunohistochemical staining,and its relationship with clinical indexes was analyzed.The TCGA database was further excavated to explore the effects of overexpression of TMIGD1 on the invasion,migration and adhesion of gastric cancer cells through Transwell,Western Blotting and cell adhesion rate experiment.Results TMIGD1 was lowly expressed in gastric cancer tissues,and the proportion of mucinous adenocarcinoma and lymphatic vessel invasion in the TMIGD1 positive group were lower than those in the TMIGD1 negative group(P<0.05).Overexpression of TMIGD1 could significantly inhibit the invasion and migration of gastric cancer cell line of AGS and BGC803,and improve cell adhesion(P<0.05).Conclusion TMIGD1 is lowly expressed in gastric cancer tissues.Overexpression of TMIGD1 can inhibit cell invasion and migration by promoting gastric cancer cell adhesion.
