中心组合设计效应面法优化枳壳中柚皮苷的提取工艺

摘要 目的:测定不同提取工艺参数下枳壳水提物中柚皮苷的含量,找到其制备的最优工艺。方法:使用Design Expert8.0软件,工艺参数选择提取时间、提取循环、水量三个,考察指标为柚皮苷含量;含量测定使用高效液相色谱法,色谱条件为Waters Xbridge C18色谱柱(柱长25cm,直径0.46mm,粒径5μm),流动相为乙腈-水(20:80),流速1.0mL/min,检测波长283nm,柱温30℃;Design Expert 8.0软件共生成20组参数,所制备20批样品均进行含量测定。结果:根据20批样品测定结果可知,当提取时间为45min、提取循环为2次、水量为枳壳饮片体积30倍时,水提物中柚皮苷含量最高,为1.6768mg/mL(平均)。结论:本方法快速、准确,能够更加有效率地对枳壳水提物的制备工艺进行优化。 Objective:To determine the content of naringin in water extract of Fructus Aurantii under different extraction parameters,and to find the optimal preparation process.Methods:Using Design Expert 8.0 software,extraction time,extraction cycle and water amount were selected as the technological parameters,and the content of naringin was investigated as the index.HPLC was performed on a Waters XBridge C18 column(column length 25 cm,diameter 0.46 mm,particle size 5μm) with mobile phase consisted of acetonitrile-water(20:80) at the flow rate of 1.OmL/min.The detection wavelength was 283 nm,and the column temperature was 30℃.A total of 20 groups of parameters were generated by Design Expert 8.0 software,and 20 batches of samples were prepared for content determination.Results:According to the determination results of 20 batches of samples,when the extraction time was 45 min,the extraction cycle was 2 times,and the volume of Fructus aurantii decoction pieces was 30 times,the content of naringin in the water extract was the highest,which was 1.6768 mg/mL(average).Conclusion:This method is rapid,accurate and can optimize the preparation process of water extract of Fructus Aurantii more efficiently.
