摘要
目的观察苦瓜复方胶囊对四氧嘧啶诱导的糖尿病模型小鼠血糖水平的影响。方法将正常小鼠随机分为对照组、苦瓜复方胶囊A高剂量组和苦瓜复方胶囊B高剂量组,每组15只,分别比较空腹血糖水平;将四氧嘧造模成功小鼠随机分为模型组、胶囊A高、中、低剂量组(1.2,0.4,0.2 g·kg^(-1))和胶囊B高、中、低剂量组(1.2,0.4,0.2 g·kg^(-1)),每组13只;给药方案为1日2次,连续30 d,必要时延长至45 d;进行糖耐量试验,并在给药后15 d、30 d、45 d测定空腹血糖,比较各组的血糖水平。结果苦瓜复方胶囊A和B对正常小鼠空腹血糖水平均无影响;给药30 d后,胶囊A高、中、低剂量组和胶囊B中、低剂量组的空腹血糖值与模型组比较有显著性降低(P<0.05);胶囊B高剂量组无显著性差异,当给药延长至45 d,空腹血糖值与30 d相比无显著性差异(P>0.05);在糖耐量试验中,胶囊A高、中、低剂量组与模型组比较,其血糖曲线下面积显著减少(P<0.01);胶囊B中、低剂量组与模型组比较,其血糖曲线下面积亦显著减少(P<0.05)。结论苦瓜复方胶囊A和B对四氧嘧啶诱导的糖尿病小鼠模型均有一定的降糖作用;胶囊A的降糖效果优于胶囊B,但需进一步毒性试验比较两者的毒性大小。
OBJECTIVE To observe the hypoglycemic effect of Compound KuGua Capsule(CKC)on the diabetic mice induced by alloxan.METHODS Normal mice were randomly divided into control group,high-dose CKC capsule A group(CKC-A)and high-dose CKC capsule B group(CKC-B),with 15 mice in each group.Diabetic mice induced by alloxan were randomly divided into Model group,CKC-A high,medium,and low dose goups(1.2,0.4,0.2 g·kg^(-1))and CKC-B high,medium,and low dose groups(1.2,0.4,0.2 g·kg^(-1)),with 13 animals in each group;the dosing regimen was 2 times per day for 30 consecutive days,extending to 45 days if necessary.The glucose tolerance test was carry out,and the fasting blood glucose was measured and compared at 15 days,30 days,and 45 days after the administration.RESULTS Both CKC-A and CKC-B had no effect on fasting blood glucose in normal mice.After 30 days of intragastric administration,the fasting blood glucose values of the CKC-A high,medium and low dose groups and of the CKC-B medium and low dose groups were significantly lower than that of the model group(P<0.05),while the CKC-B high dose group had no significant difference.Compared with 30 day s,CKC-B administration for 45 days had no significant difference in the fasting blood glucose level(P>0.05).In the glucose tolerance test,compared with the model group,AUC of CKC-A high,medium,and low dose groups and of CKC-B medium and low dose groups were significantly reduced(P<0.01).CONCLUSION CKC-A and CKC-B have certain hypoglycemic effects on diabetic mice.The hypoglycemic effect of CKC-A is better than that of CKC-B,but further toxicity tests are needed to compare the toxicity of these two.
