酶解辅助膜技术分离延胡索中生物碱的工艺研究 被引量:1

摘要 目的考察酶解辅助提取结合膜分离技术在延胡索中生物碱提取分离中的应用,优选出生物碱的最佳提取分离工艺条件。方法采用正交试验,分别考察热水浸提条件和酶解条件对延胡索中生物碱提取率的影响;使用不同截留分子量的超滤膜进行超滤,研究其对透过率、出膏率和生物碱含量的影响。结果按料液比1∶30加入蒸馏水,调节pH值为5,再加入0.1%纤维素酶,在50℃保温1.5 h,补加醋酸至浓度为3%,然后升温至70℃,提取2 h,过滤,生物碱含量可达8 mg·g^(-1);提取液使用截留分子量为1KDa的超滤膜超滤生物碱含量为4.8%;超滤液使用截留分子量为500Da纳滤膜浓缩6倍,生物碱的质量浓度提高3倍。结论酶解辅助膜技术提取分离延胡索中生物碱简便可行,能有效提高生物碱的提取率和分离效果。 OBJECTIVE The optimum extraction and separation conditions of alkaloids were selected by enzymatic assisted extraction combined with membrane separation technology.METHODS The effects of hot water extraction conditions and enzymatic hydrolysis conditions on the extraction rate of alkaloids were investigated by orthogonal test;Studying the effects of ultrafiltration membranes with different retained molecular weights on transmittance,paste yield and alkaloid content were studied.RESULTS The solid-liquid ratio was 1∶30,the pH value was 5,the cellulase concentration was 0.1%,the temperature was kept at 50℃ for 1.5 h,the acetic acid was added to the concentration of 3%,then the temperature was raised to 70℃ for 2 h,the alkaloid content was up to 8 mg·g^(-1);The extraction solution was ultrafiltered with an 1 kDa ultrafiltration membrane,and the alkaloid content was 4.8%;The ultrafiltrate was concentrated 6 times with a 500 Da nanofiltration membrane,and the mass concentration of alkaloids was increased 3 times.CONCLUSION Enzymatic hydrolysis assisted membrane technology is simple and feasible and it can effectively improve the extraction rate and separation effect of alkaloids.
