UPLC法同时测定雷公藤药材中4种生物碱的含量

摘要 目的建立超高效液相色谱法同时测定雷公藤药材中雷公藤春碱、雷公藤晋碱、雷公藤定碱和雷公藤次碱的含量,研究不同产地雷公藤药材中这4种生物碱的含量差异情况。方法采用BEH C_(18)(100 mm×2.1 mm,1.7μm)色谱柱分离,甲醇-水为流动相,梯度洗脱,流速0.2 mL·min^(-1),检测波长230 nm,柱温30℃,进样量1μL。结果雷公藤春碱、雷公藤晋碱、雷公藤定碱、雷公藤次碱4种成分在相应的范围内,呈良好的线性关系(r≥0.9999);各成分的平均加样回收率(n=6)在99.51%~99.61%之间,RSD<小于2%。其中,湖北黄石产(批号:A)的雷公藤中这4种生物碱含量均最高。结论该法可同时测定雷公藤药材中4种生物碱含量,分析速度快,溶剂用量少,重复性好,为雷公藤药材的质量评价提供参考。另外不同产地的雷公藤药材中4种生物碱的含量差异较大,在临床用药时应引起重视。 OBJECTIVE To establish a method simultaneously determine wilfortrine,wilforgine,wilfordine and wilforine in Tripterygium wilfordii Hook.f.by UPLC,and to study the difference of the content of four alkaloids in Triptergium wilfordii from different producing areas.METHODS The separation was performed on BEH C_(18)(100 mm×2.1 mm,1.7μm)column with the mobile phase consisted of methanol-water with flow rate of 0.2 mL·min^(-1) in gradient elution mode.The detection wavelength was set at 230 nm;the column temperature was 30℃;and the injection volume was 1μL.RESULTS The content of wilfortrine,wilforgine,wilfordine,and wilforine in the corresponding linear ranges showed good linear relationships(r≥0.9999);the average recovery ranged from 99.51% to 99.61% with RSD less than 2%,respectively.The content of four alkaloids in Tripterygium wilfordii Hook f.from Huangshi city,Hubei Province(Batch:A)were all the highest.CONCLUSION The UPLC method can be used for the simultaneous determination of 4 kinds of alkaloids in Tripterygium wilfordii with the fast analysis speed,small amount of solvent and good repeatability,which provides a reference method for quality assessment of Tripterygium wilfordii Hook.f..The content of 4 kinds of alkaloids in Tripterygium wilfordii Hook.f.from different producing areas vary greatly,which should be taken into account in clinic.
