摘要
目的建立北川凹叶厚朴的HPLC指纹图谱分析方法,为整体控制和评价北川凹叶厚朴的质量提供依据,评价北川凹叶厚朴从药材到饮片的整体质量变化情况。方法采用Waters e2695 HPLC,Agilent ZORBAX SB-C_(18)(4.6×250 mm,5μm)色谱柱;流动相为乙腈-0.1%磷酸水,梯度洗脱;检测波长为230 nm,以厚朴酚为参照物,对北川15批厚朴药材和对应药材制备的饮片进行指纹图谱分析。结果建立北川厚朴的HPLC指纹图谱共有模式,标定了5个共有峰,15批厚朴药材的相似度在0.975~0.998之间,15批厚朴饮片的相似度在0.981~0.999之间。结论该指纹图谱方法简便、准确、重复性好。可专属性地研究厚朴的指纹图谱,为有效地控制北川凹叶厚朴药材及饮片的内在质量提供科学依据。
OBJECTIVE To establish an HPLC fingerprint analysis method of Magnolia officinalis in Beichuan County,provide a basis for the overall control and evaluation of the quality of Magnolia officinalis in Beichuan County,and evaluate the overall quality change of Magnolia officinalis in Beichuan County from medicinal materials to decoction pieces.METHODS Using waters e2695 HPLC,Agilent Zorbax sb-C_(18)(4.6×250 mm,5μm)chromatographic column,the mobile phase is acetonitrile-0.1% phosphoric acid water,gradient elution,and the detection wavelength is 230 nm;With magnolol as the reference material,the fingerprints of 15 batches of Magnolia officinalis and the prepared pieces of corresponding medicinal materials in Beichuan were analyzed.RESULTS To establish a common mode of HPLC fingerprint of Magnolia officinalis,and five common peaks were calibrated.The similarity of 15 batches of Magnolia officinalis was 0.975-0.998,and the similarity of 15 batches of Magnolia officinalis decoction pieces was 0.981-0.999.CONCLUSION The fingerprint method is simple,accurate and reproducible.The fingerprint of Magnolia officinalis can be studied specifically to provide a scientific basis for effectively controlling the internal quality of Magnolia officinalis and its decoction pieces in Beichuan.
