摘要
目的长链非编码RNA(long non-coding RNA,lncRNA)在肿瘤发生中的作用日益到重视。本研究着眼于lncRNA H19,研究其在卵巢癌中的表达和功能。方法搜集本科室30例卵巢癌组织及癌旁组织标本,利用实时荧光定量PCR方法检测卵巢癌组织标本及癌旁组织H19的表达情况,并分析H19表达量与患者疾病进展指标的关系。通过RNA干扰技术沉默H19的在卵巢癌SKOV3细胞中的表达,CCK8法检测细胞增殖,流式细胞仪检测细胞凋亡。结果在30例患者组织标本中,相比于癌旁组织,卵巢癌组织H19表达量明显上调,且其高表达与患者FIGO分期和减瘤满意度(残余灶大小)呈正相关,而与年龄、病理分级、病理类型及术前CA125升高水平无明显相关性。进一步研究发现,相比于正常卵巢癌SKOV3细胞,沉默了H19表达的SKOV3细胞增殖速度减低,凋亡率升高。结论LncRNA H19具有促进卵巢癌恶性细胞学行为作用。
Objective Recently the role of long non-coding RNAs(lncRNAs)in carcinogenesis has been highlighted.In this study,we investigated the expression and functional role of lncRNA H19 in ovary cancer Methods The ovary cancer tissue samples as well as the adjacent normal tissues were collected from 30 patients,the expression of H19 in specimens of the ovary cancer tissues and the adjacent normal tissues was measured by real-time fluorescence quantitative PCR(qPCR).The relationship between the expression of H19 and the disease progress indicators of patients were also analyzed.Expression of H19 in ovarian cancer SKOV3 cells was silenced by RNA interference(RNAi).Cell proliferation was assessed by cell counting Kit-8 and cell metastasis and cell apoptosis rate was observed by flow cytometry analyze.Results Our results demonstrated that among 30 cases of tissue samples,in contrast to adjacent normal tissues,the expression of H19 in ovarian cancer tissue were up-regulated significantly.And it was analyzed that high expression of H19 was associated with these prognostic factors such as patients’FIGO stage and surgical satisfaction,but there was no significant correlation with age,pathological grade,pathological type and pre-operative serum CA125 expression level.With further investigation,we find that the proliferation rate H19-silenced SKOV3 OG cells was decrease,compared with that of the normal SKOV3 OG cells,and the apoptosis rate was increase.Conclusion lncRNA H19 may contribute to the malignant cell behavior of ovarian cancer.
