左氧氟沙星治疗放射性口腔黏膜炎的实验研究

Experimental study of levofloxacin on the treatment of radiation-induced oral mucositis

目的 探究左氧氟沙星(Levofloxacin,LVFX)对放射性口腔黏膜炎(radiation-induced oral mucositis,ROM)的治疗作用,从而为ROM提供更多的临床用药可能。方法 C57BL/6J小鼠头颈部接受X线放射源17 Gy一次性照射,1%甲苯胺蓝染色观察大体溃疡情况,HE染色观察舌组织病理学特征,Ki-67免疫组化检测角化上皮细胞的增殖情况。结果 与照射对照组相比,LVFX给药后的小鼠舌溃疡在大体(t=3.759,P=0.0094)、溃疡长度(t=3.132,P=0.0074)、上皮厚度(q=3.797,P=0.0012)方面都有明显改善,角化上皮细胞增殖明显(q=2.723,P=0.0420),以舌背最为显著(t=4.227,P=0.0165)。结论 LVFX照后连续给药可以有效减轻ROM的严重程度,可能对ROM有一定的治疗作用。

OBJECTIVE We investigated the therapeutic effects of levofloxacin(LVFX) on radiation-induced oral mucositis(ROM) aiming to provide more possibility of LVFX application in clinical settings.METHODS We subjected C57BL/6J mice to 17 Gy irradiation on their head and neck,after which we observed general ulcer on their tongue by 1% toluidine blue staining.HE staining and Ki-67 immunohistochemistry were also applied to explore the histopathological characteristics and detect the proliferation of keratinized epithelial cells.RESULTS Comparing with the irradiation control group,the tongue ulcer of mice treated with LVFX was significantly improved in gross(t=3.759,P=0.0094),ulcer length(t=3.132,P=0.0074) and epithelial thickness(q=3.797,P=0.0012),and the keratinized epithelial cells proliferated obviously(q=2.723,P=0.0420),especially in the dorsum of tongue(t=4.227,P=0.0165).CONCLUSION Continuous administration of LVFX after irradiation could effectively alleviate the severity of ROM and may have a certain therapeutic effect on ROM.
