Shh、Smo、Gli1在弥漫性大B细胞淋巴瘤中的表达及临床意义 被引量:2

摘要 目的探讨弥漫性大B细胞淋巴瘤(DLBCL)组织中音猬因子(Shh)、平滑受体蛋白(Smo)、神经胶质瘤相关癌基因同源蛋白1(Gli1)的表达及其临床意义。方法采用免疫组织化学SP法检测58例DLBCL组织标本及20例淋巴结反应性增生组织标本中Shh、Smo、Gli1的表达情况,分析Shh、Smo、Gli1表达与DLBCL临床病理特征的关系。结果Shh、Smo、Gli1在DLBCL组织中的阳性率分别为34.5%(20/58)、41.4%(24/58)、29.3%(17/58),均高于在淋巴结反应性增生组织中的阳性率[分别为5.0%(1/20)、5.0%(1/20)、0.0%(0/20)],差异均有统计学意义(P<0.05)。DLBCL组织中Shh、Smo、Gli1的表达均与DLBCL患者的Ann-Arbor临床分期有关(P<0.05),且分期越晚(Ⅲ+Ⅳ期),Shh、Smo、Gli1的阳性率越高。Shh的表达还与国际预后指数(IPI)评分有关(P<0.05),Smo的表达还与骨髓侵犯有关(P<0.05),而Gli1的表达还与IPI评分、B症状有关(P<0.05),但三者的表达与患者的年龄、性别,以及乳酸脱氢酶(LDH)、β2微球蛋白(β2-MG)、C反应蛋白(CRP)水平无关(P>0.05)。Spearman相关分析显示,DLBCL组织中Shh、Smo、Gli1之间的表达呈正相关(P<0.05)。结论Shh、Smo、Gli1在DLBCL组织中高表达,可能参与肿瘤的进展,有望为DLBCL的病因学研究及治疗提供理论依据。 Objective To investigate the expressions of sonic hedgehog(Shh)and smoothened(Smo),gliomaassociated oncogene homolog 1(Gli1)in diffuse large B-cell lymphoma(DLBCL)tissues and their clinical significance.Methods The expressions of Shh,Smo and Gli1 were detected in 58 cases of DLBCL tissue samples and 20 cases of lymph node reactive hyperplasia tissue samples by using immunohistochemistry SP method.The correlation between Shh,Smo and Gli1 expressions with the clinicopathological characteristics was analyzed.Results The positive rates of Shh,Smo and Gli1 in DLBCL tissues were 34.5%(20/58),41.4%(24/58)and 29.3%(17/58),respectively,which were significantly higher than 5.0%(1/20),5.0%(1/20)and 0.0%(0/20)in lymph node reactive hyperplasia tissues,and the differences were statistically significant(P<0.05).The expressions of Shh,Smo and Gli1 in DLBCL tissue were related to the Ann-Arbor clinical stage in the DLBCL patients(P<0.05),moreover the later the stage(Ⅲ+Ⅳ),the higher the positive rates of Shh,Smo and Gli1.The Shh expression was also related to the international prognostic index(IPI)score(P<0.05),the Smo expression was related to the bone marrow involvement(P<0.05),and the Gli1 expression was also related to the IPI score and B symptoms(P<0.05),but their expressions had no correlation with the age,sex,LDH,β2-MG and CRP levels(P>0.05).The Spearman correlation analysis showed that the expressions in DLBCL tissue showed the positive correlation among Shh,Smo and Gli1(P<0.05).Conclusion Shh,Smo and Gli1 are highly expressed in DLBCL tissue,which may be involved in the tumor progression and is expected to provide the theoretical basis for etiological research and treatment of DLBCL.
