摘要
目的:发现潜在可用于保湿功效评价体外研究新靶点。方法:通过GEO数据库收集正常人体皮肤数据和因皮肤保湿功能异常引起的与疾病相关的基因数据,利用GEO2R对两者基因数据进行差异分析,得到潜在与皮肤保湿相关的差异基因;再利用STRING关联丝聚蛋白基因(Filaggrin,FLG)与其他差异基因,在GeneCards数据库进行GO/KEGG富集分析,并将这些基因的生物学功能与得到的信号通路关联,阐释潜在靶基因的现实意义。结果:与丝聚蛋白基因强相关的基因有FGF1、KLK5和CASP14等。结论:FGF1、KLK5以及CASP14可作为潜在靶点化妆品保湿功效评价体外研究,具有进一步验证开发的前景。
Objective:Aiming to find potential new targets in vitro for moisturizing efficacy evaluation. Methods:Normal human skin gene data and gene data related to diseases caused by abnormal skin moisturizing function were collected through GEO database,the difference analysis between them was analyzed by GEO2R to obtain different genes;STRING was used to associate FLG and differential genes. GO and KEGG enrichment analysis were carried out with GeneCards database. The biological functions of these genes are associated with the obtained signal pathways to explain the practical significance of potential target genes related to skin moisturizing. Results:FGF1,KLK5 and CASP14 genes were strongly related to FLG gene. Conclusion:Potential targets as FGF1KLK5 and CASP14 are thought to be prospective for the evaluation of moisturizing efficacy of cosmetic study in vitro which are required for further validation and development.
