菌黄保肠合剂对脓毒症大鼠肠道菌群及血清炎症因子的影响研究 被引量:2

摘要 目的研究菌黄保肠合剂对脓毒症大鼠肠道菌群及血清炎症因子的影响。方法以110只SD大鼠为研究对象,10只为假手术组,100只采用盲肠结扎穿孔法制作脓毒症大鼠模型,分模型组、阳性对照组、菌黄保肠合剂低、中、高剂量组,阳性对照组予双歧杆菌粉(5 mg/g),中药组予菌黄保肠合剂低(1.22 mg/g)、中(6.10 mg/g)及高(12.2 mg/g),假手术组及模型组予等量灭菌注射用水,每日2次,灌胃给药。治疗3 d后记录各组大鼠存活率,采用16SrDNA测序法对大鼠结肠组织中大肠杆菌、双歧杆菌及乳酸杆菌含量进行定量分析,采用免疫比浊法检测大鼠血清降钙素原(PCT)水平,采用ELISA法检测大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平。结果菌黄保肠合剂组大鼠存活率明显高于阳性对照组(P<0.05)。与模型组比较,阳性对照组及菌黄保肠合剂低、中、高剂量组大鼠结肠组织中大肠杆菌含量明显减少,双歧杆菌和乳酸杆菌含量明显增加(P<0.05);与阳性对照组比较,菌黄保肠合剂低、中、高剂量组大鼠结肠组织中大肠杆菌含量减少,双歧杆菌和乳酸杆菌含量增加(P<0.05)。与模型组比较,阳性对照组及菌黄保肠合剂低、中、高剂量组大鼠血清PCT、TNF-α及IL-6表达水平明显降低(P<0.05);与阳性对照组比较,菌黄保肠合剂低、中、高剂量组大鼠血清PCT、TNF-α及IL-6表达水平降低(P<0.05)。菌黄保肠合剂各剂量组间大鼠存活率、肠道菌群(大肠杆菌、双歧杆菌、乳酸杆菌)及血清炎症因子(PCT、TNF-α、IL-6)等指标相比较,差异无统计学意义(P>0.05)。结论菌黄保肠合剂能调节脓毒症所致的肠道菌群紊乱,降低血清炎症因子水平,降低脓毒症的死亡率,从而发挥治疗脓毒症的作用。 Objective:To study the effects of Junhuang Baochang Decoction on intestinal flora and serum inflammatory factors in septic rats.Methods:110 SD rats were used as the research object,10 as the sham operation group and 100 as the sepsis rat model by cecal ligation and perforation.They were divided into model group,positive control group and low,medium and high dose groups of Junhuang Baochang Decoction,The positive control group was given Bifidobacterium powder(5 mg/g),twice a day.The traditional Chinese medicine group was given Junhuang Baochang Decoction low(1.22 mg/g),medium(6.10 mg/g)and high(12.2 mg/g),twice a day.The sham operation group and model group were given the same amount of sterile water for injection.After 3 days of treatment,the survival rate of rats in each group was recorded.The contents of Escherichia coli,Bifidobacterium and Lactobacillus in rat colon were quantitatively analyzed by 16SrDNA sequencing.The level of serum PCT was detected by immunoturbidimetry and the level of TNF-αand IL-6 was detected by ELISA.Results:The survival rate of rats treated with Junhuang Baochang Decoction was significantly higher than that in the positive control group(P<0.05).Compared with the model group,the contents of Escherichia coli in colon tissue of positive control group and low,medium and high dose groups of Junhuang Baochang Decoction decreased significantly,and the contents of Bifidobacterium and Lactobacillus increased significantly(P<0.05).Compared with the positive control group,the contents of Escherichia coli in colon tissue of rats in low,medium and high dose groups of Junhuang Baochang Decoction decreased(P<0.05),and the contents of Bifidobacterium and Lactobacillus increased(P<0.05).Compared with the model group,the serum levels of PCT,TNF-αand IL-6 in the positive control group and low,medium and high dose groups of Junhuang Baochang Decoction were decreased significantly(P<0.05).Compared with the positive control group,the serum levels of PCT,TNF-αand IL-6 in low,medium and high dose groups of Junhuang Baochang Decoction were decreased(P<0.05).The survival rate,intestinal flora(Escherichia coli,Bifidobacterium,Lactobacillus)and serum inflammatory factors(PCT,TNF-α,IL-6)of rats in each dose group of Junhuang Baochang Decoction were no significant differences(P>0.05).Conclusion:Junhuang Baochang Decoction can regulate the disorder of intestinal flora caused by sepsis,reduce the level of inflammatory factors and reduce the mortality of sepsis,so as to play a role in the treatment of sepsis.
