摘要
对采集自山东省青岛地区的硬毛粗盖孔菌野生子实体进行组织分离,并对该菌株进行系统鉴定、生物学特性、驯化栽培及抗肿瘤活性研究。结果表明,分离菌株YM11的ITS序列与硬毛粗盖孔菌C.trogii(MG779615.1)有最高相似性,为98%。硬毛粗盖孔菌菌丝生长的最适碳源为葡萄糖,最适氮源为酵母粉,最适温度为25℃,最适初始p H为6.5。利用A配方(木屑75%、棉籽壳10%、麸皮10%、葡萄糖4%、石膏1%,含水量60%~65%)结合管理模式实现硬毛粗盖孔菌的高产栽培,每袋产量达到1.2 kg,生物学效率达到58%。从栽培子实体提取到的粗多糖能抑制MCF-7、Hela和Hep-3B肿瘤细胞,其半数抑制浓度分别为2.43 mg·mL^(-1)、1.37 mg·mL^(-1)、2.14 mg·mL^(-1),具有广谱高效的抗肿瘤活性。研究结果可为该菌的进一步开发利用提供重要参考。
A wild fruit body of Coriolopsis trogii,collected from Qingdao,Shandong province,was isolated.In addition,systematic identification,biological characteristics,domesticated cultivation and antitumor activity of the strain were studied.The results show that,the ITS sequence of isolated strain YM11 was 98%similar to C.trogii(MG779615.1),the optimal carbon source was glucose,the optimal nitrogen source was yeast powder,the optimal temperature was 25℃,and the optimal initial pH was 6.5.Using formula A(wood chips 75%,cottonseed shell 10%,bran 10%,glucose 4%,gypsum 1%,water content 60%-65%)combined with management mode to realize high-yield cultivation of C.trogii.The yield reached 1.2 kg per bag and the biological efficiency reached 58%.The crude polysaccharides from cultivated fruit body could inhibit McF-7,Hela and HEP-3B tumor cells,and the half inhibitory concentrations were 2.43 mg·mL^(-1),1.37 mg·mL^(-1) and 2.14 mg·mL^(-1),respectively,it has broad-spectrum and high-efficiency antitumor activity.The result can provide important reference for further development and utilization of C.trogii.
