摘要
为探究不同处理条件对桑树花粉活力的影响,本研究以‘冀桑2号’、‘华桑’、‘路杨’及‘物45’四个桑树品种为研究对象,采用碘-碘化钾(I2-KI)法测定花粉活力,观察其在不同制备条件(制备温度:19℃、25℃和32℃;制备时间:1 d和7 d)、不同贮存温度(-80℃,-20℃和4℃)下的活力。实验表明,不同的制备条件和贮存温度对四个桑树品种的花粉活力均存在影响,在不同温度制备条件下,‘冀桑2号’与‘路杨’在25℃培养条件下,花粉活力最高,‘华桑’与‘物45’在19℃培养条件下,花粉活力最高;在相同的培养温度下,同一品种培养7 d的活力低于培养1 d的活力,其中19℃培养条件下的‘华桑’花粉活力最高为95.26%。四个桑树品种花粉的生活力在经过贮存一定时间后,与制备后的初始活力相比均存在不同程度的活力的下降;四个桑树品种在不同贮存温度条件下,随着储藏温度越低,花粉保存活力越高,在-80℃储藏60 d后,‘华桑’有活力的花粉最高为87.4%。综上所述,‘华桑’花粉在19℃温度下制作及-80℃保存处理后,花粉活力最高。本研究探索桑花粉的制备与贮存条件,有助于解决因气候、地域不同对杂交带来的影响,为桑树杂交育种以及生产实践以供参考依据。
In order to explore the effects of different treatments on the vitality of mulberry pollen,this study took four mulberry tree species(’Jisang2’,’Huasang’,’Luyang’and’Wu45’)as the experiment materials.The subjects were tested for vitality by the I2-KI method,and observed the vitality under different treatment(Temperature:19℃,25℃and 32℃;time:1 d and 7 d)and different storage temperatures(-80℃,-20℃and 4℃).The results have shown that different preparation conditions and storage temperatures have effects on the pollen vigor of the four mulberry varieties.Under different temperature preparation conditions,’Jisang2’and’Lu yang’had the highest pollen vigor at25℃,while’Huasang’and’Wu45’had the highest pollen vigor at 19℃.Under the same culture temperature,the vigor of the same variety after 7 days of cultivation was lower than the vigor of 1 day,among which the pollen vigor of’Huasang’under 19℃cultivation conditions was the highest pollen vigor of 95.26%.The viability of the pollen of the four mulberry varieties will decrease to varying degrees compared with the initial vigor after treatment of storage time.The four mulberry varieties under different storage temperature conditions,the lower the storage temperature,The higher the pollen preservation viability,after 60 d storage at-80℃,the highest viable pollen of’Huasang’was 87.4%.In summary,the pollen vigor of’Huasang’pollen is the highest after being prepared at 19℃and stored at-80℃.This study explored the preparation and storage conditions of mulberry pollen,which helps to solve the impact of climate and geographical differences on hybridization and provides reference for mulberry hybrid breeding and production practices.
