摘要
Aux/IAA(Auxin/Indole-3-Acetic Acid)家族是生长素信号的早期响应基因家族之一,广泛参与调控植物的生长发育过程。薄壳山核桃(Carya illinoinensis)作为一种重要经济树种,对其Aux/IAA家族基因的鉴定具有重要意义。本研究利用生物信息学方法,在薄壳山核桃全基因组中鉴定出37个Aux/IAA家族基因,亚细胞定位发现其全部成员位于细胞核。根据其与拟南芥Aux/IAA基因的系统发育关系,将37个薄壳山核桃Aux/IAA基因分为A、B两组。多重序列比对分析发现薄壳山核桃Aux/IAA蛋白具有4个保守结构域,分别为I-IV。顺式作用元件分析发现所有薄壳山核桃Aux/IAA基因启动子区域都具有响应激素的作用元件。根据薄壳山核桃Aux/IAAs在嫁接愈合中的表达模式,挖掘出4个高表达基因,它们可能在嫁接愈合过程中起重要作用。本研究将有助于阐明薄壳山核桃Aux/IAAs在嫁接愈合过程中的功能并筛选候选基因。
Aux/IAA(Auxin/Indole-3-Acetic Acid),one of primary/early auxin response genes family,is involved in regulating numerous aspects of plant growth and development.The identification of Aux/IAA genes in pecan,an important economic plant,is great significant for the study of auxin regulation mechanism.In this study,a total of 37 Aux/IAA genes were identified in the pecan genome using bioinformatics method,all of which were localized in the nucleus.Based on the phylogenic relationship with Arabidopsis Aux/IAAs,the 37 pecan Aux/IAA were categorized into two major groups(A and B).Multiple sequence alignment analysis showed that there were four typical conserved domains(I-IV)in Aux/IAA proteins of pecan.The cis-acting elements analysis revealed that all pecan Aux/IAAs had cis-acting elements in response to hormones in the promoter regions.According to the expression patterns of pecan Aux/IAA genes during graft healing,four genes with high expression were identified,which may play important roles in the formation of graft union in pecan.This study will be helpful to elucidate the function of pecan Aux/IAAs in the process of graft healing and find candidate genes.
