摘要 哺乳动物雷帕霉素靶蛋白复合物2(mTORC2)是由哺乳动物雷帕霉素靶蛋白(mTOR)为主要成分组成的复合体,mTOR是细胞生长和代谢的中央控制器,可调控细胞生长和代谢。mTORC2可通过作用于多个下游靶点调控多条信号通路,参与不同疾病的发生发展,在心血管疾病中具有重要作用。mTORC2参与自噬、免疫炎症、血管收缩以及线粒体稳态的调控,其表达下调可抑制代偿性肥厚,加速病理性心肌肥厚的发展;Rictor基因沉默以及mTORC2功能缺失导致心脏损害、寿命缩短甚至胚胎死亡;而在衰老过程中,激活mTORC2可诱导自噬并保持心脏功能。 Mammalian target of rapamycin complex 2(mTORC2)is a complex composed of mammalian target of rapamycin(mTOR),and mTOR is a central controller of cell growth and metabolism which regulates cell growth and metabolism.mTORC2 can regulate multiple signaling pathways by acting on multiple downstream targets and participate in the occurrence and development of different diseases,thus playing an important role in cardiovascular diseases.mTORC2 is involved in the regulation of autophagy,immune inflammation,vasoconstriction and mitochondrial homeostasis,which can down-regulate and inhibit compensatory hypertrophy and accelerate the development of pathological cardiac hypertrophy.Silencing of Rictor gene and loss of mTORC2 function lead to heart damage,shortened lifespan and even embryo death.Activation of mTORC2 induces autophagy and maintains cardiac function during aging.
