摘要
目的:研究光明盐-4对碘乙酰胺(IA)诱导功能性消化不良(functional dyspepsia,FD)大鼠的胃肠道运动、血清胃泌素(Gastrin,GAS)、胃动素(Motilin,MTL)及胃饥饿素(Ghrelin)水平的影响。方法:10日龄新生SD大鼠72只随机分为正常组、模型组、光明盐-4低、中、高剂量组和阳性对照组(n=12)。自10日龄起正常组每日灌胃2%蔗糖,其余组在2%蔗糖中加入0.2 mL 0.1%碘乙酰胺(IA)持续6 d。各组大鼠随后进入成年期(8周龄)后光明盐-4低、中、高剂量组给药量分别为0.3、1.5、3.0 g·(kg·d)^(-1),阳性对照组给予莫沙必利混悬液30 mg·kg^(-1),给药3周,测试行为、内脏指数、3 h进食量和胃肠运动功能及血清MTL、GAS及Ghrelin水平。结果:与正常组比较,模型组的大鼠3 h摄食总量、胃排空和小肠推进均明显减少(P<0.05),血清GAS(P<0.05)、Ghrelin水平均降低;与模型组相比,光明盐-4各组大鼠的3 h进食量、胃排空率和小肠推进率均有不同程度的增加(P<0.05);血清GAS、Ghrelin水平均显著升高(P<0.05)。结论:蒙药光明盐-4可以促进IA诱导型FD大鼠的胃肠运动功能,提高血清GAS水平,此结论可能是其治疗碘乙酰胺诱导FD大鼠的机制之一。
Objective:To study the effects of Molor Dabos-4 on gastrointestinal motility,serum gastrin(GAS),motilin(MLT)and ghrelin levels in rats with iodoacetamide(IA)induced functional dyspepsia(FD).Methods:Seventy-two 10-day-old neonatal SD rats were randomly divided into normal control group,model group,Molor Dabos-4 low-dose,medium-dose and high-dose groups and positive control group(n=12).From the 10th day of age,the normal control group was given intragastrically 2% sucrose every day,and the other groups were given 0.2 mL 0.1%I A in 2% sucrose for 6 days.When the rats in each group entered adulthood(8 weeks old),the Molor Dabos-4 low-dose,medium-dose and high-dose groups were given 0.3,1.5,3.0 g·(kg·d)^(-1),and the positive control group was given 30 mg·kg^(-1) Mosapride suspension for 3 weeks.The behavior,visceral index,3 h food intake,gastrointestinal motion function,serum MTL,GSA and ghrelin levels were measured.Results:Compared with the normal group,the 3 h food intake,gastric emptying rate and intestinal propulsive rate in IA group were significantly reduced(P<0.05),the serum GAS and Ghrelin levels were decreased(P<0.05).Compared with the IA group,the 3 h food intake,gastric emptying rate and intestinal propulsive rate in the Molor Dabos-4 group were increased with vary degrees(P<0.05),and the serum GAS and ghrelin levels were significantly increased(P<0.05).Conclusion:Mongolian medicine Molor Dabos-4 can promote gastrointestinal function and increase serum GAS level in IA induced FD rats,which may be one of its mechanisms in the treatment of iodoacetamide induced FD.
