摘要
利用小麦和玉米进行远缘杂交诱导小麦产生单倍体,再通过幼胚离体培养和人工诱导染色体加倍获得双单倍体(Double haploid,DH)植株,是构建小麦遗传育种群体的重要途径之一,能够很大程度的缩短育种进程。本文报道了矮抗58×苏隆128的F_(1)代个体与甜玉米远缘杂交后形成的单倍体胚经胚拯救后形成单倍体幼苗,再利用秋水仙碱人工诱导单倍体幼苗染色体加倍来获得纯合双单倍体的过程,最终获得156个单株的双单倍体遗传群体。本研究旨在为江淮地区小麦单倍体规模化育种提供技术支撑,并培育出性状优良的小麦新品种。
It is one of the important ways to construct wheat genetic breeding population,which can greatly shorten the breeding process,to induce wheat haploid by the distant hybridization between wheat and maize,and then to obtain double haploid(DH)plants by in vitro culture of young embryos and artificial chromosome doubling.In this paper,haploid seedlings were formed from haploid embryos of F1 generation of AK 58×Sulong 128,which were crossbred with sweet maize from a distant distance.The chromosomes of haploid seedlings were artificially induced by colchicine to double to obtain homozygous dihaploid seedlings.Two dihaploid plants were finally obtained.The aim of this study was to provide technical support for large-scale breeding of wheat haploid in Jianghuai region and to cultivate new wheat varieties with good characters.
