摘要
目的探讨长链非编码RNA(LncRNA)小核仁RNA宿主基因20(SNHG20)在前列腺癌组织中的表达及其对前列腺癌细胞增殖、迁移、侵袭和凋亡的影响。方法根据细胞处理方法将DU145细胞分为对照组(Control组)、过表达对照质粒组(Lnc-con组)、过表达组(Lnc SNHG20组)、沉默对照组(si-con组)和沉默表达组(si-SNHG20组)。实时荧光定量聚合酶链反应检测Lnc SNHG20 mRNA表达水平;噻唑蓝(MTT)法检测前列癌细胞24h、48h、72h的增殖率;培养细胞24h后Transwell检测细胞迁移率,Western blot检测迁移相关蛋白表达水平;流式细胞术检测细胞凋亡率,Western blot检测凋亡相关蛋白表达水平。结果相较于癌旁组织和人前列腺正常上皮细胞RWPE-1,前列腺癌组织和人前列腺癌细胞PC3、DU145、LNCaP中SNHG20的表达水平显著升高(P<0.05);SNHG20的表达水平与前列腺癌患者的年龄无关,与肿瘤大小、病理学分级、TMN分期和转移相关。过表达SNHG20促进细胞周期蛋白D1(Cyclin D1)、基质金属蛋白酶-2(MMP-2)的表达,抑制半胱氨酸蛋白酶-3(Caspase-3)的表达,促进细胞增殖、迁移和侵袭,抑制细胞凋亡;沉默SNHG20后,Cyclin D1、MMP-2表达受抑制,Caspase-3表达增加,抑制细胞增殖、迁移和侵袭,促进细胞凋亡。结论LncRNA SNHG20在前列腺癌组织中高表达,沉默SNHG20可抑制前列腺癌细胞增殖、迁移和侵袭,并促进细胞凋亡,可为前列腺癌的早期诊断、预防和治疗提供新标志物和新靶点。
Objective To investigate the expression of long non-coding RNA(LncRNA)small nucleolar RNA host gene 20(SNHG20)in prostate cancer tissues and its effects on proliferation,migration,invasion and apoptosis of prostate cancer cells.Methods According to cell treatment methods,DU145 cells were divided into Control group,Lnc-con group(transfected with Lnc-con),the Lnc SNHG20 group(transfected with Lnc SNHG20),the si-con group(transfected with si-con),and the si-SNHG20 group(transfected with si-SNHG20).Lnc SNHG20 mRNA expression was detected by real-time fluorescence quantitative polymerase chain reaction.MTT assay was used to detect the proliferation rate of prostolic cancer cells at 24 h,48 h and 72 h.Transwell was used to detect cell mobility and Western blot was used to detect the expression levels of migration-related proteins.The apoptosis rate was detected by flow cytometry and the expression level of apoptosis-related proteins was detected by Western blot.Results Compared with paracancerous tissues and human prostate normal epithelial cells RWPE-1,the expression of SNHG20 in prostate cancer tissues and human prostate cancer cells PC3,DU145,LNCaP were significantly increased(P<0.05).The expression level of SNHG20 was not correlated with the age of prostate cancer patients,but with tumor size,pathological grade,TMN stage and metastasis.Overexpression of SNHG20 promoted the expression of Cyclin D1 and matrix metalloproteinase-2(MMP-2),inhibited the expression of Caspase-3,promoted cell proliferation,migration and invasion,and inhibited apoptosis.After SNHG20 silencing,the expression of Cyclin D1 and MMP-2 were inhibited,and the expression of Caspase-3 was increased,which inhibited cell proliferation,migration and invasion and promoted cell apoptosis.Conclusions LncRNA SNHG20 is highly expressed in prostate cancer tissues.Silencing SNHG20 can inhibit the proliferation,migration and invasion of prostate cancer cells and promote apoptosis,providing a new marker and target for the early diagnosis,prevention and treatment of prostate cancer.
