黄精对原发性痛经大鼠药效作用初步研究 被引量:4

摘要 目的建立原发性痛经大鼠模型,初步研究黄精对原发性痛经大鼠的药效作用。方法采用大鼠雌激素与缩宫素联合法建立原发性痛经大鼠模型。通过观察大鼠的扭体反应,判断原发性痛经大鼠模型是否建立成功;ELISA法检测大鼠血清中6-keto-PGF_(1)α和β-EP的含量以及子宫组织中PGF_(2)α和PGE_(2)的含量;离体组织器官恒温灌流系统测定子宫肌张力。结果与正常组相比,模型组大鼠扭体潜伏时间缩短,扭体次数明显增加,表明原发性痛经大鼠模型建立成功。与模型组相比,各治疗组大鼠血清中6-keto-PGF_(1)α、β-EP的含量显著提升,大鼠子宫组织中PGF_(2)α的含量明显降低,PGE_(2)的含量显著提升,子宫肌张力明显下降。HE染色结果表明,治疗组大鼠子宫上皮细胞空泡病变基本消失,充血现象明显改善。结论黄精对原发性痛经大鼠具有良好的治疗效果,其可能与大鼠体内PGE_(2)、PGF_(2)α等因子有关。 Objective To determine the pharmacodynamic effect of Polygonatum on primary dysmenorrhea in rat models.Methods Estrogen and oxytocin were combined to establish primary dysmenorrhea model in rats.By observing the writhing response of rats,we decided whether the model of primary dysmenorrhea was established.ELISA was used to detect the serum concentration of 6-keto-PGF_(1)αandβ-EP,and that in the uterine tissue PGF_(2)αand PGE_(2).The tension of uterine muscle was measured by constant temperature perfusion system.Results Compared with the normal group,the writhing latency of the rats in the model group was shortened,and the number of writhings were significantly increased,indicating successful establishment of rat model of primary dysmenorrhea.Compared with the model group,in the treatment group 6-keto-PGF_(1)αandβ-EP in the serum were significantly increased,the PGF_(2)αcontent in the rat uterus decreased,while the PGE_(2)content was increased,and the uterine muscle tension was decreased.HE staining showed that the vacuolation of uterine epithelial cells in the treatment group basically disappeared,and the congestion was significantly improved.Conclusion Polygonatum has a good therapeutic effect on rats with primary dysmenorrhea,which may be related to factors such as PGE_(2)and PGF_(2)αin rats.
