基于网络药理学的山豆根保肝作用药效物质筛选及实验验证 被引量:1

Screening and validation of hepatoprotective substances of Sophorae Tonkinensis Radix et Rhizoma based on network pharmacology

摘要 目的采用网络药理学方法探讨山豆根治疗肝病的药效物质,并进行初步验证。方法检索中药系统药理学数据库以及TCMSP分析平台,筛选山豆根中的化学成分。通过GeneCards、OMIM、PharmGkb以及DrugBank数据库获取肝病相关疾病靶点,运用Cytoscape 3.8.2软件进行网络构建;并基于微流控芯片及酶联免疫吸附法对预测的药效成分展开验证。结果山豆根与肝病的靶点取交集后共获得170个共同靶点,其中槲皮素富集的靶点最多。验证结果显示,槲皮素、山柰酚、异鼠李素对L-02肝损伤模型、HSC-T6、HepG-2细胞的侵袭作用明显,可使TNF-α和IL-6分泌显著降低(P<0.05)。结论基于网络药理学方法实现山豆根药效物质的筛选及预测,为山豆根治疗肝病的科学解释提供实验数据。 Objective To determine the pharmacodynamic substances of Sophorae Tonkinensis Radix et Rhizoma for liver diseases based on network pharmacology,and to verify by experiments preliminarily.Methods The pharmacological database of traditional Chinese medicine system and TCMSP platform were retrieved to screen the chemical components in Sophorae Tonkinensis Radix et Rhizoma.The targets of liver diseases were obtained through GeneCards,OMIM,PharmGkb and DrugBank databases,and the network was constructed by Cytoscape 3.8.2 software.The predicted pharmacodynamic components were verified by microfluidic chip and enzyme linked immunosorbent assay.Results Totally 170 common targets were obtained after the intersection of Sophorae Tonkinensis Radix et Rhizoma and liver disease,of which quercetin enriched the most targets.Quercetin,kaempferol and isorhamnetin had obvious invasion on L-02 liver injury model,HSC-T6 and HepG-2 cells.TNF-α and IL-6 secretion were decreased significantly (P<0.05).Conclusion Based on the network pharmacology,pharmacodynamic substances of Sophorae Tonkinensis Radix et Rhizoma are screened and predicted to provide experimental data for the scientific interpretation of mountain bean root in the treatment of liver disease.
