摘要
目的基于P物质(SP)和P-糖蛋白(P-gp)探讨黄芪多糖(APS)温敏凝胶对小鼠放射性皮炎的作用及其机制。方法小鼠随机分为正常对照组、模型组、APS温敏凝胶组和空白温敏凝胶对照组,通过10 Gy X线连续照射5 d制备放射性皮炎小鼠模型;采用RT-PCR考察APS温敏凝胶对SP mRNA和炎性因子mRNA的影响,采用Western blot法测定皮肤P-gp的表达;采用HE染色、甲苯胺蓝染色和CD31免疫荧光染色评价APS温敏凝胶对小鼠放射性皮炎病理学的影响。结果与模型组比较,0~5 d时APS温敏凝胶组SP mRNA升高,10 d和15 d时APS温敏凝胶SP、IL-6、IL-10和IL-12 mRNA均显著降低,P-gp的表达升高。结论APS温敏凝胶可增加SP和P-gp的表达,降低炎症因子的表达,有效缓解小鼠放射性皮炎。
Objective To determine the effect and mechanism of thermosensitive gel of Astragalus polysaccharides(APS)on radioactive dermatitis in mice based on substance P(SP)and P-glycoprotein(P-gp).Methods The mice were randomly divided into a normal control group,a model group,an APS thermosensitive gel group and a blank thermosensitive gel group.The mice model of radioactive dermatitis was established by 10 Gy X-ray irradiation for 5 days.RT-PCR was used to determine the mRNA expression of SP and inflammatory factors.P-gp expression was evaluated by Western blot.HE staining,toluidine blue staining and CD31 immunofluorescence staining were used to evaluate the effect of APS thermosensitive gel on the pathology of radioactive dermatitis in mice.Results Compared with the normal control group,SP mRNA of the APS thermosensitive gel group was increased on 0~5 d,and mRNA of inflammatory factors decreased on 10 d and 15 d,and the expression of P-gp increased.Conclusion APS thermosensitive gel can increase the expression of SP and P-gp,decrease the expression of inflammatory factors,and effectively alleviate radioactive dermatitis in mice.
