摘要
目的:通过使用中药网络药理学研究方法,探讨射干麻黄汤治疗支气管哮喘的作用机制。方法:通过中药、疾病公共数据库,收集射干麻黄汤有效活性成分及其作用于疾病的作用靶点;分别构建中药成分与疾病基因的蛋白互作网络,并筛选射干麻黄汤治疗支气管哮喘作用的关键靶点;对其进行生物学过程富集分析与通路富集分析,探究其可能的作用机制。结果:检索TCMSP数据库获得成分1494个,根据ADME筛选原则纳入144个;收集成分作用靶点1658个;检索GeneCards数据库,共收集与支气管哮喘相关的靶点1832个;从网络中筛选关键靶点115个;对其进行分析可知其主要涉及576种生物学过程,111条信号通路,其中包括转录调控、RNA聚合酶Ⅱ启动子转录的正调控、病毒过程、肽基酪氨酸自磷酸化、fcε受体信号通路、信号转导、肽基酪氨酸磷酸化、蛋白质磷酸化、刺激性C型凝集素受体信号通路等生物学过程;PI3K/AKT、神经营养素、FoxO、Erbb、趋化因子、焦点粘附、TNF等信号通路。结论:射干麻黄汤中化学成分多样,治疗支气管哮喘途径广泛,分子作用机制复杂,具有抑制炎症、改善气道重塑、提高免疫等多方面治疗效果。中医药治疗支气管哮喘理论可行,值得推广。
Objective:To explore the mechanism of Shegan Mahuang Decoction in treating bronchial asthma by using the method of network pharmacology of traditional Chinese medicine.Methods:Effective active ingredients of Shegan Mahuang Decoction and their action targets on diseases were collected through public databases of traditional Chinese medicine and diseases;protein interaction networks between Chinese medicine ingredients and disease genes were constructed,and key targets of Shegan Mahuang Decoction in the treatment of bronchial asthma were screened;enrichment analysis of biological processes and pathways was carried out to explore its possible mechanism of action.System.Results:A total of 1494 components were obtained from TCMSP database,and 144 were included according to ADME screening principle;1658 targets were collected;1832 BA-related targets were collected from GeneCards database;115 key targets were screened from the network;576 biological processes and 111 signaling pathways were identified,including transcriptional regulation and RNA aggregation.Positive regulation of enzyme II promoter transcription,viral process,peptide tyrosine autophosphorylation,fc-epsilon receptor signaling pathway,signal transduction,peptide tyrosine phosphorylation,protein phosphorylation,stimulating C-type lectin receptor signaling pathway and other biological processes;PI3K/AKT,neurotrophin,FoxO,Erbb,chemokine,focal adhesion,TNF and other signaling pathways.Conclusion:Shegan Mahuang Decoction has a variety of chemical constituents,a wide range of ways to treat bronchial asthma,and complex molecular mechanisms.It can inhibit inflammation,improve airway remodeling,and improve immunity.The theory of traditional Chinese medicine in treating bronchial asthma is feasible and worth popularizing.
