基于网络药理学探讨青风藤穿山龙药对治疗强直性脊柱炎的作用机制 被引量:1

The Active Mechanism of Sinomenii acutum-Dioscoreae nipponicae Makino Herb Pairs in the Treatment of Ankylosing Spondylitis Based on Network Pharmacology

摘要 本研究基于网络药理学探讨青风藤穿山龙药对治疗强直性脊柱炎(AS)的作用机制。通过TCMSP数据库筛选青风藤和穿山龙的有效化学成分,在PharmMapper服务器获取成分的蛋白质靶点信息,在DrugBank、GeneCards及OMIM数据库获得疾病的靶点基因,借助String平台构建交集基因PPI网络,在Metascape网络完成GO及KEGG通路富集分析。结果显示,共获得药物有效成分16个,有效成分靶点基因297个,疾病靶点基因816个,交集基因56个。PPI网络分析得到关键靶点基因是ALB、IGF1、ESR1、AR、MMP2和IL2。GO富集分析得到GOBP 739条,GOCC 19条和GOMF 79条。KEGG富集分析得到74条通路,主要信号通路为类固醇激素生物合成、补体和凝血级联、foxo信号通路、AMPK信号通路、PI3K-Akt信号通路和T细胞受体信号通路。青风藤穿山龙药通过多靶点和多通路共同起到调节免疫反应、抑制炎症过程及调控骨细胞活性等作用治疗强直性脊柱炎。 The study investigated the mechanism of action of Sinomenii acutum-Dioscoreae nipponicae Herb Pairs in the treatment of ankylosing spondylitis(AS)based on network pharmacology.The effective chemical constituents of Sinomenii cautum and Dioscoreae nipponicae were screened through the TCMSP database,the protein target information of the ingredients was obtained from PharmMapper server,the target genes of diseases were obtained from DrugBank,GeneCards and OMIM database,the PPI network of intersection genes was constructed with the help of the String platform.GO enrichment analysis and KEGG pathway enrichment analysis were completed in Metascape network.The results showed that a total of 16 active drug ingredients,297 active component target genes,816 disease target genes and56 intersection genes were obtained.PPI network analysis showed that the key target genes were ALB,IGF1,ESR1,AR,MMP2 and IL2.GO enrichment analysis revealed 739 GOBP,19 GOCC and 79 GOMF.KEGG enrichment analysis revealed 74 pathways,including steroid hormone biosynthesis,complement and coagulation cascade,FOXO signaling pathway,AMPK signaling pathway,PI3K-Akt signaling pathway and T cell receptor signaling pathway.The drug can regulate the immune response,inhibit the inflammatory process and regulate the activity of osteocytes to treat ankylosing spondylitis through multi-target and multi-pathway.
