摘要
目的:检测microRNA‐4513(miR‐4513)在胃癌中的表达并其相关的临床意义。方法:选取41例胃癌患者的手术标本以及胃癌细胞系HGC27和MGC803,采用实时荧光定量-聚合酶链反应(RT-PCR)方法检测胃癌组织和细胞中miR-4513的表达,分析miR-4513表达与患者临床病理特征的关系,COX回归模型分析影响患者生存期的因素,Kaplan-Meier生存曲线分析miR-4513不同表达水平患者的生存率。结果:HGC27和MGC803胃癌细胞中miR-4513 mRNA表达显著高于GES正常胃粘膜上皮细胞,胃癌组织中miR-4513表达显著高于癌旁组织,差异均有统计学意义(P<0.01)。miR-4513的表达与TNM分期、淋巴结转移及远处转移有关(P<0.05)。多因素分析显示miR-4513可以作为一个独立表示预后的因子,miR-4513高表达组患者的总生存率低于低表达组,差异有统计学意义(P<0.01)。结论:miR-4513可能在胃癌的发生发展中具有一定的促进作用,miR-4513高表达组患者可能预后不良。
Objective:To examine the expression of miR-4513gene in human gastric carcinoma(GC)and explore its clinical significance.Medthods:The expression of miR-4513 in 41 GC tissues and cells was examined by using quantitative real-time PCR.The relationship between miR-4513 expression and clinicopathological features of patients was analyzed.COX regression model was used to analyze the factors affecting the survival of patients,and Kaplan-Meier survival curve was used to analyzethe survival rate of patients with different levels of miR-4513 expression.Results:miR-4513 was highly expressed in gastric cancer cells HGC27 and MGC803 than that in normal gastric mucosa epithelial cells GES.miR-4513 expression in gastric carcinoma tissues was significantly higher than that in adjacent tissues,and the difference was statistically significant(P<0.01).The expression of miR-4513 was correlated with TNM stage,lymph node metastasis and distant metastasis of gastric cancer(P<0.05).Multifactorial analysis showed that miR-4513 may be regarded as a prognostic factorfor survival.The overall survival rate of miR-4513 high-expressed group was lower than that of miR-4513 low-expressed group(P<0.001).Conclusion:miR-4513 may play a role in promoting the development of gastric carcinoma,and patients with high expression of miR-4513 may have a poor prognosis.
