羌活胜湿汤治疗强直性脊柱炎的网络药理学分析 被引量:2

摘要 目的:基于网络药理学探讨羌活胜湿汤治疗强直性脊柱炎的作用机制。方法:在中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)筛选羌活胜湿汤组方中药的化学成分,以口服利用度(oral availability,OB)≥30%、类药性(drug likeness,DL)≥0.18为条件进行筛选,得到羌活胜湿汤的活性成分,并通过TCMSP数据库进一步预测所有活性成分的相关靶点。在人类基因数据库(GeneCards)和GeneBank数据库搜索强直性脊柱炎的相关靶点,采用Venn2.1.0在线平台获取药物靶点和疾病靶点的交集靶点,并使用STRING在线数据库构建交集靶点的蛋白相互作用关系。采用Metascope数据库对交集靶点进行基因本体(gene ontology,GO)功能富集分析和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。结果:共获得到羌活胜湿汤活性成分61个,活性成分相关靶点265个;在GeneCards和GeneBank数据库搜索强直性脊柱炎靶点共1672个,活性成分靶点与疾病相关靶点映射后得到交集靶点26个。GO功能富集分析得到GO条目620条,其中生物过程475条、分子功能124条、细胞组分21条。KEGG通路富集分析共得到28条信号通路,主要涉及类固醇激素生物合成通路、NOD样受体信号通路、Toll样受体信号通路等。结论:羌活胜湿汤可通过多成分、多通路、多靶点治疗强直性脊柱炎。 Objective:To explore the mechanism of Qianghuo Shengshi Decoction in treatment of ankylosing spondylitis based on network pharmacology.Methods:The traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)was used to screen the chemical constituents of Qiang Huo Sheng Shi Decoction.Screening was carried out under the conditions of oral availability(OB)≥30%and drug likeness(DL)≥0.18 to obtain the active ingredients of Qianghuo Shengshi Decoction,and further predict the correlation of all active ingredients through the TCMSP database.Search the related targets of ankylosing spondylitis in the human gene database(GeneCards)and GeneBank database,use the Venn2.1.0 online platform to obtain the intersection targets of drug targets and disease targets,and use the STRING online database to construct the protein-protein interaction relationship.The Metascope database was used to perform gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis for the intersection targets.Results:A total of 61 active ingredients and 265 active ingredient-related targets were obtained from Qianghuo Shengshi Decoction;a total of 1672 ankylosing spondylitis targets were searched in the GeneCards and GeneBank databases,and the active ingredient targets were mapped with disease-related targets.26 intersection targets were obtained.GO functional enrichment analysis yielded 620 GO items,including 475 for biological processes,124 for molecular functions,and 21 for cellular components.After KEGG enrichment,P<0.05 and FDR<0.05 were used as the criteria for screening,and a total of 28 signaling pathways were obtained,mainly involving steroid hormone biosynthesis pathway,NOD-like receptor signaling pathway,and Toll-like receptor signaling pathway.Conclusion:Qianghuoshengshi Decoction can treat ankylosing spondylitis through multi-component,multi-channel and multi-target.
