摘要
目的测定新疆绝经后女性2型糖尿病(T2DM)患者的DKK1 rs1373004、rs1528877位点基因多态性并分析其与骨代谢的关系,为T2DM合并OP机制的进一步明确及防治提供理论依据。方法收集石河子地区2016年10月—2017年10月绝经后女性136例,分为4组:对照组(A组)、骨量异常组(B组)、T2DM组(C组)、T2DM伴骨量异常组(D组)。研究女性绝经之后的年龄、年限与BMI等的关系。并在此过程中对血糖、糖化血红蛋白(HbA1c)以及三酰甘油(TG)等生化指标进行检测。结果A组的FPG及HbA1c较C、D组降低,C、D组的TG、BMD(L1-4)和BMD降低,差异有统计学意义(P<0.05)。组间基因型及等位基因分布频率:rs1373004位点,与A组相比,B组及D组的分布差异有统计学意义(P<0.05);与B组相比,D组的分布差异有统计学意义(P<0.05)。rs1528877位点,与A组相比,C组和D组的分布差异有统计学意义(P<0.05)。多元线性回归分析:rs1373004位点基因多态性、TG与BMD(L1-4)成正相关。结论在新疆石河子地区绝经后女性中,两位点基因多态性与糖、骨代谢有关。rs1373004位点基因多态性、TG的升高是BMD(L1-4)升高的保护因素。
Objective To determine the gene polymorphisms of DKK1 rs1373004 and rs1528877 in postmenopausal women with type 2 diabetes(T2DM)in Xinjiang and analyze their relationship with bone metabolism.To provide a theoretical basis for the further clarification and prevention of the mechanism of T2DM combined with OP.Methods A total of 136 postmenopausal women in Shihezi area from October 2016 to October 2017 were collected.Divided into four groups:control group(group A),abnormal bone mass group(group B),T2DM group(group C),T2DM with abnormal bone mass group(group D).To study the relationship between postmenopausal women's age,years and BMI.During this process,biochemical indicators such as blood glucose,glycosylated hemoglobin(HbA1c)and triglycerides(TG)were detected.Results The FPG and HbA1c of group A was lower than that of group C and D,TG,BMD(L1-4)and BMD of group C and D decreased,and the differences were statistically significant(P<0.05).The frequency of genotype and allele distribution between groups:rs1373004 locus,the distribution of group B and D compared with group A,the difference was statistically significant(P<0.05);the distribution of group D compared with group B,the difference was statistically significant(P<0.05).rs1528877 locus,the distribution of group C and D compared with group A,the difference was statistically significant(P<0.05).Multiple linear regression analysis:rs1373004 gene polymorphism,TG and BMD(L1-4)were positively correlated.Conclusion In postmenopausal women in Shihezi area of Xinjiang,two-point gene polymorphisms are related to sugar and bone metabolism.rs1373004 gene polymorphism and elevated TG are protective factors for elevated BMD(L1-4).
