慢性丙肝患者血清半乳糖凝集素-4和白细胞介素-27的表达变化分析

Analysis of changes in the expression of serum galactoglucose-4 and interleukin-27 in patients with chronic hepatitis C infection

目的 探究血清半乳糖凝集素-4(GAL4)和白细胞介素-27(IL-27)在慢性丙型肝炎患者中的表达变化及临床意义。方法 选取39例慢性丙肝患者和30例健康人作为研究对象;酶联免疫吸附试验测定血清GAL4和IL-27浓度;荧光定量PCR测定外周血单个核细胞(PBMC)中GAL4和IL-27 mRNA的表达水平;记录研究对象的临床资料。Spearman相关分析法分析GAL4和IL-27与临床参数间的相关性。结果 慢性丙肝患者血清GAL4和IL-27以及PBMC中GAL4和IL-27mRNA均显著高于对照组(P<0.05);血清GAL4及GAL4 mRNA与AST和HCV RNA均呈显著负相关性。血清IL-27和IL-27 mRNA与HCV RNA呈显著负相关性,与ALB均呈显著正相关性。结论 外周血GAL4和IL-27水平在慢性丙肝患者中显著升高,并与病毒载量密切相关,或可成为反映慢性丙肝患者病毒活跃程度的标志物。

Objective To investigate the changes in the expression of serum galactose lectin-4(GAL4) and interleukin-27(IL-27) in patients with chronic hepatitis C infection and their clinical significance.Methods Thirty-nine patients with chronic hepatitis C infection and 30 healthy subjects were selected as study subjects.Serum GAL4 and IL-27 concentrations were measured by enzyme-linked immunosorbent assay.The expression levels of GAL4 and IL-27 mRNA in peripheral blood mononuclear cells(PBMC) were determined by fluorescence quantitative PCR.The clinical data of the study subjects were also recorded.Spearman’s correlation analysis was performed to analyze the relationship between GAL4,IL-27 and clinical parameters.Results It was showed that GAL4 and IL-27 were significantly higher in patients with chronic hepatitis C infection than thoes in controls(P<0.05).Serum GAL4 and GAL4 mRNA were negatively correlated with AST and HCV RNA.Serum IL-27 and IL-27 mRNA showed significant negative correlations with HCV RNA and significant positive correlations with ALB.Conclusion Circulating GAL4 and IL-27 levels were significantly elevated in patients with chronic hepatitis C infection and were closely correlated with viral load,which may be markers of viral activity for patients with chronic hepatitis C infection.
