C57BL/6小鼠和C57BL/6 FMR1基因敲除小鼠行为学及针刺长强穴治疗后的对比研究

摘要 目的:通过避暗实验、跳台实验和矿场实验来研究C57BL/6小鼠和C57BL/6 FMR1基因敲除小鼠的行为学差异,并通过针刺长强穴治疗后进行行为学对比。方法:通过胚胎复苏得到3只雌性杂合的C57BL/6 FMR1基因敲除小鼠后与C57BL/6小鼠进行繁殖,最终得到纯合的雌性和雄性C57BL/6 FMR1基因敲除小鼠各10只进行行为学实验。结果:在避暗实验的第3天,WT(野生型)组小鼠的潜伏期大于KO(基因敲除型)组,WT组小鼠的电击次数低于KO组,且均存在显著性差异(P<0.05);在跳台实验中,WT组小鼠的潜伏期和错误次数均低于KO组小鼠(P<0.05);在旷场实验中,WT组小鼠的平均速度明显高于KO组小鼠(P<0.01),WT组小鼠在中央区域停留的时间高于KO组小鼠(P<0.05)。通过针刺长强穴治疗后,在避暗实验中,长强穴组小鼠的潜伏期高于非经非穴组、电击次数低于非经非穴组,且都存在显著性差异(P<0.05);跳台实验中的潜伏期和旷场实验中的平均速度以及中央区域停留时间,长强穴组均高于非经非穴组(P<0.05)。结论:在行为学实验中,KO组小鼠主要受到FMR1基因敲除影响,使KO组小鼠趋避性增加,对环境的认知能力及探索性下降,焦虑程度增强,其学习记忆能力和运动能力明显低于WT组小鼠,在针刺长强穴治疗后,通过避暗实验、跳台实验和旷场实验等行为学实验的对比实验表明:C57BL/6 FMR1基因敲除小鼠是理想的脑瘫、脑卒中后遗症等疾病康复的实验动物模型,并且针刺治疗对其有显著的改善效果。 Objective:The behavioral differences between C57BL/6 mice and C57BL/6 FMR1 knockout mice were studied by stepthrough experiment,step-down experiment and open field test.Methods:Three female heterozygous C57BL/6 FMR1 knockout mice were obtained by embryo resuscitation,and then propagated with C57BL/6 mice.Finally,10 homozygous female and 10 male C57BL/6 FMR1 knockout mice were obtained for behavioral experiments.Results:On the 3rd day of the dark avoidance experiment,the latency of mice in the WT(wild-type)group was greater than that in the KO(knockout)group,and the number of electric shocks in the WT group was lower than that in the KO group,and there were significant differences(P<0.05).In the step-downexperiment,the latency of mice in KO group was significantly higher than that in WT group(P<0.05),and the number of errors in WT group was significantly lower than that in KO group(P<0.05).In the open field test,the average speed of mice in WT group was significantly higher than that in KO group(P<0.01).The time of staying in the central area in WT group was higher than that in KO group(P<0.05).After acupuncture at DU1 acupoint,in the stepthrough experiment,the latency of DU1 acupoint group was higher than that of non-acupoint group and non-acupoint group,and the number of electric shocks was lower than that of non-acupoint group group(P<0.05);In step-down experiment,the latency of DU1 acupoint was significantly higher than that of non-acupoint group(P<0.05);In the open field test,the average velocity and central area residence time of DU1 acupoint group were significantly higher than those of non-acupoint group(P<0.05).Conclusion:In the behavioral experiment,KO group mice were mainly affected by FMR1 gene knockout,which increased the avoidance,decreased the cognitive ability and exploratory ability to the environment,and increased the degree of anxiety.Their learning,memory and motor ability were significantly lower than those of WT group mice.After acupuncture at DU1 acupoint,through the comparative experiments of step-through experiment,stepdown experiment and open field experiment,it is shown that C57BL/6 FMR1 knockout mice is an ideal experimental animal model for the rehabilitation of cerebral palsy,stroke sequelae and other diseases,and has a significant improvement effect on acupuncture treatment.
