基于GEO芯片数据水稻干旱和盐胁迫共同差异表达基因分析

Analysis of Common Differentially Expressed Genes in Rice under Drought and Salt Stress based on GEO Database

摘要 干旱和盐胁迫是影响水稻生长发育的主要环境因子,为了获得两种胁迫下共同差异表达基因,选取GEO数据库干旱和盐胁迫两种处理条件下基于GPL2025平台的7个数据集,利用GEO2R进行相关差异基因分析和Venn分析法筛选干旱和盐胁迫下共同差异表达基因。结果表明:在所选数据集中,干旱和盐胁迫下共同差异表达基因共有3个,分别是编码11 kD胚胎发育晚期丰富蛋白基因LOC_Os08g23870、脱水素Rab16B样蛋白基因LOC_Os11g26780和一个未知功能蛋白基因LOC_Os11g07911;基本性质预测分析显示3个基因表达的蛋白质均具亲水性,且定位于细胞核,前两者较为稳定,后者不稳定;3个基因的启动子序列区含有丰富的脱落酸应答元件和茉莉酸甲酯元件。水稻幼苗经干旱和盐胁迫处理后,幼苗叶片和根部均可检测到LOC_Os08g23870、LOC_Os11g26780和LOC_Os11g07911不同程度的上调表达,暗示这3个基因对水稻耐受干旱胁迫和盐胁迫具有重要的生物学意义。本研究为进一步探索LOC_Os08g23870、LOC_Os11g26780和LOC_Os11g07911功能以及培育抗旱耐盐种质材料提供了理论基础。 Drought and salt stress are main environmental factors affecting the rice growth and development.In order to obtain the common differentially expressed genes under the two stresses,seven drought and salt stress-related datasets based on GPL2025 platform were selected,GEO2R was used to explore the differentially expressed genes,and Venn analysis was conducted to screen the common differentially expressed genes between drought and salt stress.The results showed that there were three common differentially expressed genes in the selected datasets,which were LOC_Os08g23870,LOC_Os11g26780 and LOC_Os11g07911,encoding the 11 kD late embryogenesis abundant protein,dehydrin Rab16B-like protein and an uncharacterized protein,respectively.Prediction analysis of basic properties suggested that three proteins encoded by the selected genes were hydrophilic and localized in the nucleus,meanwhile the former two proteins were stable,and the latter one was unstable.The promoter regions of the three genes possessed abundant abscisic acid response and methyl jasmonate elements.Additionally,the expressions of LOC_Os08g23870,LOC_Os11g26780 and LOC_Os11g07911 exhibited different up-regulations in the leaves and roots of rice seedling treated with drought and salt stress,respectively.These results indicated that the three genes implicated important biological significance for rice tolerance to drought and salt stress.This study provided a theoretical foundation for the further functional exploration of LOC_Os08g23870,LOC_Os11g26780 and LOC_Os11g07911 and cultivation of drought and salt tolerant rice germplasm.
