结直肠腺癌中沉默信息调节因子1、缺氧诱导因子-1α、突变型P53蛋白的表达及意义 被引量:3

Expressions and significances of silent information regulator 1, hypoxia-inducible factor-1α and mutant P53 proteins in colorectal adenocarcinoma

摘要 目的探讨结直肠腺癌组织中沉默信息调节因子1(SIRT1)、缺氧诱导因子-1α(HIF-1α)、突变型P53蛋白的表达水平及其临床意义。方法收集2015年3月至2021年10月山西省中医院68例经病理确诊为结直肠腺癌患者资料,采用免疫组织化学法检测SIRT1、HIF-1α和突变型P53蛋白在结直肠腺癌组织及癌旁组织中的表达情况,分析三种蛋白的相关性及其与患者临床病理特征的关系。结果68例结直肠腺癌组织和癌旁组织中,SIRT1蛋白阳性分别为38例(55.88%)、11例(16.18%)(χ^(2)=23.25,P<0.001);HIF-1α蛋白阳性分别为47例(69.12%)、5例(7.35%)(χ^(2)=54.92,P<0.001),突变型P53蛋白表达分别为41例(60.29%)、0例(0)(P<0.001)。临床分期高、有淋巴结转移患者SIRT1蛋白阳性表达率高(均P<0.05);分化程度越低,HIF-1α阳性表达率越高(P<0.05);分化程度低、有淋巴结转移患者的突变型P53阳性表达率高(均P<0.05)。SIRT1和突变型P53蛋白表达呈负相关(r_(s)=-0.38,P=0.001),HIF-1α与突变型P53蛋白表达呈正相关(r_(s)=0.56,P<0.001),SIRT1与HIF-1α蛋白表达呈负相关(r_(s)=-0.40,P=0.001)。结论SIRT1、HIF-1α、突变型P53蛋白在结直肠腺癌中均呈高表达状态,且与提示预后不良的临床病理特征具有相关性,联合检测三种蛋白可能作为结直肠腺癌诊断及预后的检测手段及治疗的新靶点。 Objective To investigate the expression levels of silent information regulator 1(SIRT1),hypoxia-inducible factor-1α(HIF-1α)and mutant P53 proteins in colorectal adenocarcinoma tissues and their clinical significances.Methods The data of 68 cases of colorectal adenocarcinoma confirmed by pathology in Shanxi Traditional Chinese Medical Hospital from March 2015 to October 2021 were collected.The expressions of SIRT1,HIF-1αand mutant P53 proteins in colorectal adenocarcinoma tissues and paracancerous tissues were determined by immunohistochemistry.The correlation among SIRT1,HIF-1αand mutant P53 proteins and their relationship with clinicopathological features of patients were analyzed.Results Among 68 colorectal adenocarcinoma tissues and paracancerous tissues,SIRT1 protein was positive in 38 cases(55.88%)and 11 cases(16.18%)(χ^(2)=23.25,P<0.001),HIF-1αprotein was positive in 47 cases(69.12%)and 5 cases(7.35%)(χ^(2)=54.92,P<0.001),and mutant P53 protein was positive in 41 cases(60.29%)and 0 cases(0)(P<0.001).The positive expression rate of SIRT1 protein was high in patients with high clinical stage and lymph node metastasis(both P<0.05);the positive expression rate of HIF-1αprotein was high in patients with poor differentiation(P<0.05);the positive expression rate of mutant P53 protein was high in patients with poor differentiation and lymph node metastasis(both P<0.05).There was a negative correlation between expressions of SIRT1 and mutant P53 proteins(r_(s)=-0.38,P=0.001);there was a positive correlation between expressions of HIF-1αand mutant P53 proteins(r_(s)=0.56,P<0.001);there was a negative correlation between expressions of SIRT1 and HIF-1αproteins(r_(s)=-0.40,P=0.001).Conclusions SIRT1,HIF-1αand mutant P53 proteins are highly expressed in colorectal adenocarcinoma and are correlated with clinicopathological features suggesting poor prognosis.Combined detection of the three proteins may be used for the diagnosis and prognosis of colorectal adenocarcinoma and serve as a new target for treatment.
