合成载小分子肽羊胎素碳酸钙/磷酸钙纳米材料的方法研究

摘要 碳酸钙/磷酸钙是脊椎动物骨骼和牙齿的主要无机成分,具有良好的生物相容性,在生物医学领域具有良好的应用前景。从羊胎儿中提取的小分子肽羊胎素具有很好的生物学作用,为进一步提高小分子肽羊胎素在皮肤中的渗透性和功效,利用生物矿化法合成载小分子肽羊胎素碳酸钙/磷酸钙纳米材料,利用扫描电镜、傅立叶红外光谱对纳米颗粒进行表征,检测其载小分子肽能力和pH的敏感性。结果表明:利用小分子肽羊胎素溶液实现Ca^(2+)、CO_(3)^(2-)、HPO_(4)^(2-)共沉淀获得小分子肽/碳酸钙/磷酸钙复合纳米颗粒同时高效载小分子肽(1.8%),纳米颗粒具有球形结构,直径小于50 nm,且具有较好的pH响应性和蛋白质持续释放能力。在兔和猫眼上试验表明载小分子肽纳米颗粒对动物眼睛无刺激性,有较好的生物安全性。这一研究为利用天然小分子肽合成钙纳米材料提供了依据,同时为进一步开发新一代小分子肽羊胎素纳米材料应用于化妆品奠定了基础。 Calcium carbonate/calcium phosphate is the main inorganic component of vertebrate bones and teeth,which has good biocompatibility and has a good application prospect in the biomedicine.Small molecular peptides extracted from caprine fetus has positive biological effect.To further improve small molecular peptides permeability and efficacy in the skin,we prepared peptided CaCO_(3)/CaHPO_(4) nanoparticles(pCCN)using a simple reaction between small molecular peptides from caprine fetus,calcium(Ca^(2+)),and carbonate(CO_(3)^(2-))/phosphate(HPO_(4)^(2-))ions.The obtained pCCN were characterized by scanning electron microscope(SEM)and fourier transform infrared(FTIR).Small molecular peptides were loaded in the pCCN with high encapsulation efficiency(1.8%).The synthesized pCCN had a spherical structure with a diameter of less than 50 nm and better pH responsiveness and protein sustained release ability.Small molecular peptides from the nanoparticles could be effectively sustained.Tests on rabbit and cat eyes showed that the nanoparticles had no irritation to animal eyes and had good biosafety.The results provide a theoretical basis for the synthesis of calcium nanomaterials using natural small molecule peptides from caprine fetus,and lay a foundation for the development of a new generation of small molecule peptide nanomaterials for cosmetics.
