桂枝茯苓胶囊对高脂去卵巢大鼠血清内皮微颗粒的影响及内皮保护作用

摘要 目的:探讨桂枝茯苓胶囊对高脂去卵巢大鼠血清内皮微颗粒(Endothelial Microparticles,EMPs)的影响和血管内皮保护的机制。方法:建立高脂去卵巢大鼠模型,将大鼠随机分为正常对照组、假手术组、模型组、阳性药物组和桂枝茯苓胶囊组。采用酶法检测大鼠总胆固醇(Total Cholesterol,TC)、三酰甘油(Triglyceride,TG)、低密度脂蛋白胆固醇(Low-density Lipoprotein Cholesterol,LDL-C)和高密度脂蛋白胆固醇(High-density Lipoprotein Cholesterol,HDL-C)的含量;采用放射免疫法检测大鼠血清中雌二醇(Estradiol,E;)含量;采用流式细胞术检测大鼠血清中EMPs水平;采用蛋白免疫印迹法(Western blot)检测大鼠主动脉雌激素受体(Estrogen Receptor,ER)蛋白表达。结果:模型组大鼠血清中TC、TG、LDL-C和EMPs水平明显升高,HDL-C和E;水平明显降低,主动脉ER蛋白表达明显下调;相较于模型组,桂枝茯苓胶囊可显著下调TC、TG、LDL-C和EMPs水平,上调HDL-C、E;和ER水平。结论:桂枝茯苓胶囊可明显降低去卵巢大鼠的血脂水平,升高大鼠血清雌激素水平,降低EMPs水平,从而抑制高脂去卵巢大鼠的血管内皮损伤。 Objective:The effects of the Guizhi Fuling capsules(桂枝茯苓胶囊)on serum endothelial microparticles in high-fat ovariectomized rats,and the vascular endothelial protection mechanism were investigated.Methods:The model of ovariectomized rats was established,and the rats were randomly divided into the normal control group,the sham operation group,the model group,the positive drug group and the Guizhi Fuling capsules group.The enzymatic method was used to detect the contents of TC,TG,LDL-C and HDL-C in serum.The radioimmunoassay technology was used to detect the density of E2 in serum.The flow cytometry technology was used to detect the level of EMPs in serum.The protein expression of ER in rat aorta was detected by Western blot.Results:The levels of TC,TG,LDL-C and EMPs in the model group were significantly increased,the levels of HDL-C and E2 in serum of rats were significantly decreased.The protein expression of ER in the aortic tissues of rat was significantly down-regulated.Compared with the drugs used in the model group,the Guizhi Fuling capsules could significantly down-regulate the levels of TC,TG,LDL-C,and EMPs,and increase the levels of HDL-C,E2 and ER.Conclusion:The Guizhi Fuling capsules can significantly reduce blood lipid levels in ovariectomized rats,increase serum estrogen levels,and reduce EMPs levels,thereby inhibiting vascular endothelial injury in high-fat ovariectomized rats.
