白藜芦醇对动脉粥样硬化大鼠脂代谢及抗氧化作用的研究 被引量:2

摘要 目的:探讨不同浓度白藜芦醇对动脉粥样硬化模型大鼠脂代谢和抗氧化的影响。方法:选取60只无特定病原体级雄性SD(Sprague-Dawley)大鼠,随机分成6组:空白组,模型组,白藜芦醇高(Res-H)、中(Res-M)、低(Res-L)(200 mg/kg、100 mg/kg、50 mg/kg)剂量组和阿托伐他汀钙片组(西药组,8 mg/kg),每组10只。采用高脂膳食联合腹腔注射维生素D;复制动脉粥样硬化大鼠模型。给药完成后,检测大鼠血清总胆固醇(Total Cholesterol,TC)、三酰甘油(Triglyceride,TG)、低密度脂蛋白胆固醇(Low Density Lipoprotein Cholesterol,LDL-C)、高密度脂蛋白胆固醇(High Density Lipoprotein Cholesterol,HDL-C),并分析动脉硬化指数(Arteriosclerosis Index,AI)。计算丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Total Superoxide Dismutase,T-SOD)、一氧化氮(Nitric Oxide,NO)含量和抗超氧阴离子(O;-)、抗羟自由基(-OH)能力以及大鼠胸主动脉油红O染色,观察其病理变化。结果:与空白组相比较,模型组大鼠脂代谢能力降低(P <0.01),AI;、AI;显著升高(P <0.01),抗氧化能力显著降低(P <0.01),油红O染色显示内膜下脂质沉积有动脉粥样硬化斑块形成。与模型组相比,白藜芦醇中、低剂量组对大鼠血脂改善及抗氧化能力不显著(P <0.05);白藜芦醇高剂量组能显著改善动脉粥样硬化大鼠血脂代谢(P <0.01),提高抗氧化能力(P <0.01),油红O染色显示内膜光滑,内皮完整,无动脉粥样硬化斑块形成。结论:高剂量白藜芦醇可以改善动脉粥样硬化大鼠血脂水平,发挥良好的抗氧化作用,抑制动脉粥样硬化的形成。 Objective: To explore the effects of different concentrations of resveratrol on lipid metabolism and antioxidation in atherosclerotic rats. Methods: 60 specified pathogen-free male SD rats were randomly divided into 6 groups, namely the blank group,the model group, the high, middle and low dose of resveratrol groups(received 200 mg/kg, 100 mg/kg, and 50 mg/kg of resveratrol respectively) and the atorvastatin calcium tablets group(received 8 mg/kg of atorvastatin calcium tablets), with 10 rats in each group.Atherosclerotic rat models were replicated with high-fat diet plus intraperitoneal injection of vitamin D;. After administration, the levels of serum TC, TG, LDL-C, and HDL-C in the rats were detected. The AI values were analyzed. The MDA, T-SOD, and NO levels were measured. The ability of anti-superoxide anion and anti-hydroxyl radical was counted. The pathological changes of rat thoracic aorta were observed by oil red O staining. Results: Compared with the blank group, the lipid metabolism ability in the model group was decreased(P<0.01), the level of AI;and AI;in the model group were increased significantly(P<0.01), the antioxidant capacity in the model group was decreased significantly(P<0.01), and the oil red O staining showed atherosclerotic plaques in lipid deposits under the intima. Compared with the model group, the middle and low dose of resveratrol groups did not obviously ameliorate blood lipid and antioxidant capacity of rats(P<0.05), while the high dose of resveratrol group could significantly ameliorate blood lipid metabolism(P<0.01), and improve antioxidant capacity of atherosclerotic rats(P<0.01). The oil red O staining showed that the intima was smooth, the endothelium was intact, and there was no atherosclerotic plaque formation. Conclusions: High-dose resveratrol can ameliorate the level of blood lipids of atherosclerotic rats, play a good role of antioxidation, and inhibit the formation of atherosclerosis.
