干制扇贝柱多糖热水辅助酶解法提取工艺优化

Optimization of Extraction Process of Polysaccharide from Dried Scallop Column by Hot Water Assisted Enzymatic Hydrolysis

摘要 以干制扇贝柱为原料,通过热水辅助酶解法提取干制贝柱多糖,利用单因素试验和正交试验得到最佳工艺条件。在料液比为1∶40,90℃热水浸提4 h,上清液经浓缩、醇沉,用木瓜蛋白酶酶解,酶解时间3 h,温度57.8℃,pH值7.1,加酶量2.0%,酶解液离心取上清液,醇沉得到扇贝多糖纯度为48.62%±0.79%。 The polysaccharide from dried scallop column was extracted by hot water assisted enzymatic hydrolysis.The optimum process conditions were obtained by single factor and orthogonal test.The supernatant was extracted with hot water at 90℃for 4 hours at the ratio of liquid to material of 1∶40.The supernatant was concentrated and alcohol precipitated,and enzymolysis was carried out with papain.The enzymolysis time was 3 hours,the temperature was 57.8℃,the pH was 7.1,and the amount of enzyme was 2.0%.The supernatant was taken from the enzymolysis heart,and the purity of scallop polysaccharide was 48.62%±0.79%.
