针灸对自然衰老围绝经期大鼠雌激素水平及单胺类神经递质的影响

Effects of acupuncture and moxibustion on estrogen levels and monoamine neurotransmitters in naturally aging perimenopausal rats

目的探讨针灸肾俞、三阴交和神门对围绝经期大鼠单胺类神经递质及雌激素水平的影响。方法将30只12~14月龄SD大鼠随机分为模型组、阳性对照组和治疗组,每组10只;另取4~6月龄SD大鼠10只作为对照组。治疗组针刺双侧肾俞、三阴交和神门,阳性对照组灌胃给予尼尔雌醇(剂量为1.5 mg/kg)。给药结束后24 h测定大鼠行为学变化;苏木素-伊红(HE)染色观察大鼠卵巢组织病理情况;采用放射免疫分析法测定大鼠血清中雌二醇(E2)、卵泡刺激素(FSH)和促黄体生成素(LH)水平;采用高效液相色谱法测定大鼠脑组织去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(HT)含量水平。结果对照组卵巢组织黄体发育良好,体积较大,可见多个成熟卵泡。模型组卵巢组织黄体数目少,成熟卵泡少。阳性对照组和治疗组卵巢萎缩现象明显改善。与模型组比较,治疗组血清E2水平明显升高(P<0.05),血清FSH和LH水平明显降低(P<0.05),脑组织NE和DA水平明显升高(P<0.05),5-HT含量水平均明显降低(P<0.05)。结论针灸肾俞、三阴交和神门能调节血清雌激素水平,并通过调节脑组织单胺类神经递质水平,改善围绝经期抑郁样行为。

Objective To investigate the effects of acupuncture Shenshu,Sanyinjiao and Shenmen on monoamine neurotransmitter and estrogen levels in perimenopausal rats.Methods Thirty SD rats aged 12 to 14 months were randomly divided into model group,positive control group and treatment group,with 10 rats in each group.Another 10 SD rats aged 4~6 months were used as control group.The treatment group was needled with bilateral Shenshu,Sanyinjiao and Shenmen,and the positive control group was given nilestriol(1.5 mg/kg)by gavage.The behavioral changes of rats were determined 24 h after administration.Hematoxylin-eosin(HE)staining was used to observe the pathological conditions of the ovarian tissues of rats.The levels of estradiol(E2),follicle stimulating hormone(FSH)and luteinizing hormone(LH)in serum of rats were determined by radioimmunoassay.The contents of norepinephrine(NE),dopamine(DA)and 5-hydroxytryptamine(HT)in rats brain were determined by HPLC.Results In the control group,the luteum of the ovary was well developed and large,and multiple mature follicles were observed.The number of luteum and mature follicles in the ovarian tissue of model group were less.Ovarian atrophy was significantly improved in positive control group and treatment group.Compared with model group,serum E2 level in treatment group was significantly increased(P<0.05),serum FSH and LH levels were significantly decreased(P<0.05),brain NE and DA levels were significantly increased(P<0.05),and 5-HT content was significantly decreased(P<0.05).Conclusions Acupuncture Shenshu,Sanyinjiao and Shenmen could regulate the level of serum estrogen and the level of monoamine neurotransmitter in brain tissue,and improve the depressive behavior in perimenopausal period.
