摘要 为探索高压蒸汽灭菌与过滤除菌的PEG干旱胁迫下对甜玉米萌发的影响,探寻最佳模拟干旱胁迫的方式,对在干旱条件下实施玉米的抗逆性试验提供指导。本试验用过滤除菌和高压蒸汽灭菌处理的5种不同浓度PEG,模拟干旱胁迫下玉米种子发芽情况,经过测定玉米种子的发芽率、发芽指数、活力指数以及幼苗含水量。对所得数据进行方差分析以及相关性分析、主成分分析、聚类分析。结果表明,高压蒸汽灭菌与过滤除菌后不同的PEG模拟干旱胁迫下对甜玉米萌发存在影响,玉米发芽率、发芽指数、活力指数以及幼苗含水量均与PEG浓度呈显著(p<0.05)负相关。经主成分分析、聚类分析结果表明,过滤灭菌、高压蒸汽灭菌的5%PEG与无菌水为轻微胁迫水平,过滤灭菌、高压蒸汽灭菌的10%的PEG与过滤灭菌15%的PEG为中度胁迫水平,过滤灭菌、高压蒸汽灭菌的20%的PEG与高温蒸汽灭菌15%的PEG为重度胁迫水平。PEG进行高压蒸汽灭菌与过滤灭菌均可用于玉米萌发过程中的干旱胁迫模拟,其中15%PEG适合用于模拟中度胁迫,20%PEG适合用于模拟重度胁迫。 Explore the direct autoclaving and filter sterilization PEG treatment to simulate the influence of sweet corn germination under drought stress,find the best way to simulate drought stress,and provide guidance for the implementation of corn stress resistance test under drought conditions.This experiment used filtration five different concentrations of PEG treated with sterilization and high-temperature steam sterilization simulated the germination of corn seeds under drought stress.The germination rate,germination index,vitality index and seedling water content of corn were determined.Perform variance analysis,correlation analysis,principal component analysis,and cluster analysis on the obtained data.The results showed that direct autoclaving and filter sterilization had different effects on sweet corn germination under different PEG simulated drought stress.The germination rate,germination index,vigor index and seedling water content of corn were significantly related to the PEG concentration(p<0.05).negative correlation.The results of principal component analysis and cluster analysis showed that filter sterilization,autoclave sterilization of 5%PEG and sterile water was a mild stress level,filter sterilization,autoclave sterilization of 10%PEG and filter sterilization 15%PEG was classified as moderate stress level,filter sterilization,high-pressure steam sterilization 20%PEG and high-temperature steam sterilization 15%PEG was classified as severe stress level.Both high-pressure steam sterilization and filter sterilization of PEG wase used to simulate drought stress during corn germination.15%PEG was suitable for simulating moderate stress,and 20%PEG was suitable for simulating severe stress.
