小绿叶止咳糖浆主要化合物鉴定及药物移行成分分析

Identification of main compounds and analysis of drug transitional components for Prospan Hustenliquid Cough Syrup

目的鉴定小绿叶止咳糖浆主要化合物,并分析药物移行成分。方法该药物50%乙腈提取液的UPLC-QTOF/MS分析采用Acquity UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8μm);流动相0.05%甲酸(或水)-乙腈,梯度洗脱;体积流量0.3 mL/min;柱温40℃;电喷雾离子源,正负离子扫描模式。结果鉴定了62个主要化合物,包括酚苷、植物激素各1个,萜类3个,氨基酸、有机酸各5个,香豆素3个,黄酮7个,皂苷34个。发现了32种药物移行成分,在大鼠肺组织中的含量高于血浆中。结论该方法准确稳定,可为小绿叶止咳糖浆质量评价和控制奠定基础。

AIM To identify main compounds in Prospan Hustenliquid Cough Syrup and to analyze drug transitional components.METHODS The analysis of 50%acetonitrile extract of this drug was performed on a 40℃thermostatic Acquity UPLC HSS T3 column(100 mm×2.1 mm,1.8μm),with the mobile phase comprising of 0.05%formic acid(or water)-acetonitrile flowing at 0.3 mL/min in a gradient elution manner,and electrospray ionization was adopted in positive and negative ion scanning mode.RESULTS Sixty-two main compounds were identified,containing one phenolic glycoside,one plant hormone,three terpenoids,five amino acids,five organic acids,three coumarins,seven flavonoids and thirty-four saponins,and thirty-two drug transitional components were found,whose contents were higher in rat lung tissue than those in plasma.CONCLUSION This accurate and stable method can lay the foundation for quality evaluation and control of Prospan Hustenliquid Cough Syrup.
