唐古特大黄根茎不同组织的红外光谱分析及药效学的对比研究 被引量:2

Infrared spectrum analysis and comparative study on the pharmacodynamics inflammatory of different tissues of Rheum Tanguticum Maxim

摘要 目的利用红外光谱分析唐古特大黄根茎不同组织中成分分布特征,同时对比研究唐古特大黄根茎不同组织的泻下及抗炎作用。方法使用溴化钾压片法对样品进行红外光谱测定。(1)将小鼠随机分为空白组、模型组、唐古特大黄根茎组、传统药材组和外皮层组。空白组灌胃蒸馏水(20 mL·kg^(-1)),其余各组灌胃复方地芬诺酯(50 g·kg^(-1))复制便秘模型。空白组与模型组灌胃空白对照液(20 mL·kg^(-1)),其余给药组分别灌胃加入碳素墨汁的唐古特大黄根茎药液、传统药材药液、外皮层药液(均为4 g·kg^(-1))。给药后观察小鼠首次排黑便时间、5 h内排便次数及测定肠道含水量,次日,灌胃1 h后,用酶联免疫吸附法(ELSA)测定小鼠结肠Na^(+)-K^(+)-ATP酶活性。(2)用二甲苯致小鼠耳肿胀法、甲醛致小鼠足肿胀法建立炎症模型,将小鼠随机分为即模型组、唐古特大黄根茎组、传统药材组和外皮层组。模型组灌胃蒸馏水(20 mL·kg^(-1)),其余给药组分别灌胃唐古特大黄根茎药液、传统药材药液、外皮层药液(均为4 g·kg^(-1)),每天1次,连续3 d。计算小鼠耳肿胀度及足肿胀度。结果不同组织唐古特大黄红外图谱没有明显差异。空白组、模型组、唐古特大黄根茎组、传统药材组和外皮层组Na^(+)-K^(+)-ATP酶的活性分别为(19.60±4.10),(42.19±4.24),(20.77±3.04),(25.56±3.65)和(35.01±2.83)μmol·h^(-1)·g^(-1),唐古特大黄根茎组、传统药材组、外皮层组与模型组比较,差异有统计学意义(P<0.05,P<0.01)。模型组、唐古特大黄根茎组、传统药材组和外皮层组的耳肿胀度分别为(5.22±1.94),(2.67±0.90),(3.20±0.98)和(3.53±1.44)mg;足肿胀度分别为(78.07±9.23),(59.70±6.67),(60.65±10.00)和(62.25±8.98)mg,唐古特大黄根茎、传统药材、外皮层组与模型组比较,差异有统计学意义(P<0.01)。结论唐古特大黄根茎不同组织所含化学成分非常相似,唐古特大黄根茎、传统药材、外皮层有明显的泻下及抗炎作用,提示唐古特大黄在产地加工过程中可不必刮去外皮。 Objective To analyze the distribution characteristics of components in different tissues of Rheum Tanguticum Maxim used infrared spectroscopy,at the same time,comparative study on the purgative and anti-inflammatory effects of different tissues of Rheum Tanguticum Maxim.Methods The potassium bromide tablet method was used to determine the sample by infrared spectrum.(1)Mice were randomly divided into blank group,model group,Rheum Tanguticum Maxim group,traditional medicine group and exodermis group.Blank group was given distilled water(20 mL·kg^(-1))by intragastric administration,and other groups were given compound diphenoxate(50 g·kg^(-1))by intragastric administration to make constipation model.Blank group and model group were given blank liquid(20 mL·kg^(-1)),and the other treatment groups were intragastrically supplemented with carbon ink Rheum Tanguticum Maxim liquid,traditional medicinal liquid,exodermis liquid(all 4 g·kg^(-1)),after administration,the time of the first black stool in mice and the times of defecation within 5 h were observed.The next day,1 h after intragastric administration,colon Na^(+)-K^(+)-ATPase activity was determined by enzyme linked immunosorbent assay(ELISA).(2)The inflammation model was established by ear swelling induced by xylene and foot swelling induced by formaldehyde.Mice were randomly divided into model group,Rheum Tanguticum Maxim group,traditional medicine group,exodermis group.Model group was given distilled water(20 mL·kg^(-1))by intragastric administration and the other treatment groups were given Rheum Tanguticum Maxim liquid,traditional medicinal liquid,exodermis liquid(all 4 g·kg^(-1)),once a day for 3 days,weighing and calculating the degree of ear swelling and foot swelling.Results There was no obvious difference in the infrared spectra of different tissues of Rheum Tanguticum Maxim.The activities of Na^(+)-K^(+)-ATPASE in blank group,model group,Rheum Tanguticum Maxim group,traditional medicine group and exodermis group were(19.60±4.10),(42.19±4.24),(20.77±3.04),(25.56±3.65)and(35.01±2.83)μmol·h^(-1)·g^(-1),Compared between Rheum Tanguticum Maxim group,traditional medicine group,exodermis group and model group can obviously shorten the defecation time of constipated mice,increase the frequency of defecation,increase the water content of large intestine,and inhibit the activity of Na^(+)-K^(+)-ATPase in colon.The above effects of Rheum Tanguticum Maxim are obviously stronger than those of traditional medicinal,the difference was statistically significant(P<0.05,P<0.01).The degree of ear swelling in model group,Rheum Tanguticum Maxim group,traditional medicine group and exodermis group were(5.22±1.94),(2.67±0.90),(3.20±0.98)and(3.53±1.44)mg;the degree of foot swelling were(78.07±9.23),(59.70±6.67),(60.65±10.00)and(62.25±8.98)mg,compared with the blank group,the Rheum Tanguticum Maxim,traditional medicine and exodermis could significantly relieve the swelling degree of auricle and foot of mice and the difference was statistically significant(P<0.01).Conclusion The chemical constituents in different tissues of Rheum Tanguticum Maxim are very similar;Rheum Tanguticum Maxim,traditional medicine and exodermis has obvious purgative and anti-inflammatory effects;it is suggested that there is no need to scrape off the skin of Rheum Tanguticum Maxim in the process of producing.
