摘要
试验设计3对引物,采用PCR产物直接测序检测BMP2基因在短毛黑水貂和咖啡水貂共252个个体中的单核苷酸多态性,分析该基因对水貂毛长性状的影响,并对序列进行了分析。结果表明,外显子1、2和3在2个水貂品种中均无多态性;内含子1有1个SNPs:C159T位点,但是和毛长性状无相关性。水貂扩增序列与雪貂的同源性为98.2%,与其它哺乳动物的同源性都在81.2%以上。研究结果表明,水貂BMP2基因外显子1、2和3序列保守性高,该区域不是影响水貂毛长的功能结构域。
Three pairs of primers were designed.Single nucleotide polymorphisms(SNPs)of BMP2 gene were detected in 252 minks by PCR amplification and sequencing techniques.The effect of this gene on the hair length of mink and its sequence were analyzed.The results showed that no polymorphism is detected in exons 1,2 and 3.The SNPs C159T is found in intron 1,but not correlated with hair length traits.The homology of the amplified sequence of mink to ferret is 98.2%,and the homology to other mammals is more than 81.2%.The results indicated that exons 1,2 and 3 of BMP2 gene are highly conserved among mammals,and may not be the functional domains that affect the hair length in minks.
