从海泥中分离的约氏不动杆菌聚磷特性研究 被引量:1

摘要 试验从海泥中筛选聚磷菌,通过生理生化和分子生物学对聚磷菌进行鉴定,利用钼酸铵分光光度法研究聚磷菌的聚磷特性,筛选高效聚磷菌,为水产养殖中生物除磷技术提供新参考。结果表明,从海泥中分离出一株聚磷菌,经生理生化特征及16S rDNA序列分析,鉴定该菌为约氏不动杆菌(Acinetobacter johnsonii);在0.1 mg/L~10 mg/L磷浓度的培养基中,该菌72 h后平均聚磷率达到73.59%;当pH 6~8、温度28~33℃条件下该菌聚磷效果最佳,聚磷率能达到70%以上;在模拟养殖水中,以不同比例加入该菌,72 h后平均聚磷率达到70.75%。研究结果表明,分离并鉴定的该株聚磷菌具有较强的聚磷效果。 The experiment screened phosphorus-accumulating bacteria from the sea mud,then identified it through physiological,biochemical and molecular biology.Using ammonium molybdate spectrophotometry,the phosphorus-accumulating characteristics of it was studied.As a result,high-efficiency phosphorus-accumulating bacteria was screened,providing a new reference for biological phosphorus removal technology.The results showed that a phosphorus-accumulating bacteria was isolated from sea mud,and it was identified as Acinetobacter johnsonii by its physiological and biochemical characteristics and 16S rDNA sequence analysis.In the medium with 0.1 mg/L~10 mg/L phosphorus density,the average phosphorus accumulation rate of the bacteria reached 73.59%after 72 h;when the pH was 6 to 8 and the temperature was 28 to 33℃,the bacteria had the best phosphorus accumulation effect,and the phosphorus accumulation rate could reach more than 70%;in simulated aquaculture water,adding the bacteria with different proportions,the average phosphorus accumulation rate reached 70.75%after 72 h.The research results show that the isolated and identified strain of phosphorus accumulating bacteria has strong phosphorus accumulating effect.
