生物信息学分析肉瘤抗原1与卵巢浆液性囊腺癌预后及免疫细胞浸润的关系

CLINICAL SIGNIFICANCE OF SAGE1 IN PRIGNOSIS AND IMMUNE INFILTRATION IN OV BASED ON BIOINFORMATICS ANALYSIS

目的应用生物信息学方法分析肉瘤抗原1(sarcoma antigen I,SAGE1)在卵巢浆液性囊腺癌(OV)中的表达,分析其与肿瘤免疫细胞浸润的关系,并探讨SAGE1在OV中的预后价值。方法通过癌症基因组图谱数据库(tissue Cancer Genome Atlas,TCGA)及基因型组织表达(genotype-Tissue Expression,GTEx)数据库分析SAGE1mRNA在OV患者及正常卵巢组织中的表达差异。Kaplan-Meier曲线及COX回归分析用于患者预后评估,通过Tumor IMmune Estimation Resource(TIMER)网站分析SAGE1表达与OV患者免疫细胞浸润的相关性。结果SAGE1mRNA表达明显高于正常卵巢组织;Kaplan-Meier和COX回归模型分析显示高表达SAGE1与浆液性卵巢癌患者不良预后相关(P=0.027),SAGE1高表达是OV患者预后的独立危险因素。SAGE1表达与多种免疫细胞及细胞表面标志物呈正相关。结论通过生物信息学的数据挖掘发现SAGE1在OV中高表达,且与OV患者的预后及免疫细胞浸润存在相关性。

Objective Bioinformatics analysis was used to explore the correlation between the expression of SAGE1 and immune infiltration in OV and to investigate the prognostic value of SAGE1 in OV.Methods The Cancer Genome Atlas(TCGA)and the Genotype-Tissue Expression(GTEx)were served as data resources.Wilcoxon rank test was performed to evaluate the expression difference of the SAGE1 between cancer tissues and normal tissues.Kaplan-Meier curve and Cox regression were used for prognosis evaluation.The Tumor IMmune Estimation Resource(TIMER)website was used to analyze the relationship between SAGE1 and immune cell infiltration in OV.Results SAGE1 mRNA was higher in OV tissues,OV patients with higer SAGE1 expression had a worse prognosis,and SAGE1 expression was a independent prognostic factor.The expression of SAGE1 was positively correlated with different immune cells or immune cell markers.Conclusion SAGE1 is highly expressed in OV patients,and its high expression was a independent prognostic factor.The expression of SAGE1 was related with the infiltration of immune cells or immune cell markers.
