摘要
目的通过生物信息学方法筛选巴雷特食管的关键基因及信号通路。方法从GEO数据库中下载GSE39491、GSE36223、GSE13083三组数据集,获得差异表达基因(|Log_(2)FC|>1,调整后的P<0.01)并绘制火山图,并筛选出差异共表达基因,使用Web Gestalt数据库对差异共表达基因进行GO和KEGG分析,并使用String和Cytoscape构成蛋白-蛋白互作网络图,筛选显著模块和关键基因。结果筛选出327个差异共表达基因,其中表达上调181个,表达下调146个,GO和KEGG分析提示基因在角质化、角化细胞分化、视黄醛脱氢酶的激活、白介素-1受体的结合、药物代谢、细胞色素P450的异生物质代谢等方面富集。利用Cytoscape筛选出的关键基因为PPL、EVPL、IVL、TGM1、PI3、DSG3、PKP1、SPRR1A、FLG、DSC3。结论关键基因与巴雷特食管的发生发展相关,有望成为其生物标志物。
Objective To screen the key genes and signal pathways in Barrett’s esophagus by bioinformatics method.Methods From the GEO database,three sets of data were download as follow,GSE39491,GSE36223,GSE13083 to obtain differentially expressed genes(| Log_(2)FC |>1,the adjustedP value<0.01),map the volcano,and screen out the differential co-expressed genes.The WebGestalt database was used to analyze the differentially co-expressed genes by GO and KEGG analysis,and the String and Cytoscape were used to construct the protein-protein interaction network map to screen the significant modules and key genes.Results 327 differentially expressed genes were selected,among which 181 were up-regulated and 146 were down-regulated.GO and KEGG analysis indicated that these genes were enriched in keratinization,keratinocyte differentiation,activation of aldehyde dehydrogenase,binding of interleukin-1 receptor,drug metabolism,and heterobascularization of cytopigment P450.The key genes screened out by Cytoscape were PPL,EVPL,IVL,TGM1,PI3,DSG3,PKP1,SPRR1 A,FLG and DSC3.Conclusion The key genes are associated with the occurrence and development of Barrett’s esophagus and may be used as biomarkers.
