摘要
激活蛋白-1(activator protein-1,AP-1)是由JUN和FOS原癌基因产物组成的二聚体DNA结合蛋白。作为转录激活因子,AP-1参与调节细胞增殖、分化、及凋亡等多种生物学过程,在肿瘤的形成过程中起着重要作用。因此,通过抑制AP-1活性来治疗癌症充满希望。本文就AP-1的结构、信号通路、与肿瘤的关系以及靶向AP-1信号通路的抗肿瘤天然产物及其结构修饰等方面作一综述。
Activator protein-1(AP-1)is a dimeric DNA binding protein composed of the products of the JUN and FOS proto-oncogenes.As a transcriptional activator,AP-1 is implicated in a variety of cellular processes,including cell proliferation,differentiation and apoptosis,and plays an important role in tumorigenesis.Therefore,treatment of cancers through the inhibition of AP-1 appears promising.This review aims to elucidate the structural characteristics of AP-1,AP-1 associated signaling pathways and the relationship between AP-1 and tumors,and to discuss the anti-tumor natural products targeting AP-1 and their structural modifications.
