产胡萝卜素酵母菌的筛选

摘要 为获得具有开发应用价值的产胡萝卜素的酵母菌株,本研究从自然界中共采集了35个土样,经过富集、涂布、划线分离、种子培养和发酵培养后,采用二甲基亚砜和丙酮以1:3的比例进行破壁提取,并对色素提取物进行紫外扫描定性、定量分析,得到7株酵母菌株符合试验要求,并筛选出胡萝卜素产量最高的菌株,编号为21-1,产量为7.42 mg/L。对此菌株进行无机盐组分单因素试验和正交试验的初步优化,优化后21-1菌株胡萝卜素产量为8.55 mg/L,相比初筛的菌种提高了15.3%。将这7株菌株产胡萝卜素的特征吸收峰与β-胡萝卜素标准品进行对比发现,其中一种含量为β-胡萝卜素。 To develop useful carotene-producing yeast strains.A total of 35 soil samples were collected from nature,after enrichment,coating,line separation,seed culture and fermentation,two methyl sulfoxide and acetone were extracted by 1:3,and the pigment extracts were analyzed qualitatively and quantitatively with high performance liquid chromatography and ultraviolet scanning,and 7 strains of yeast were conformed to the experimental requirements.After calculation,the highest yield strains were selected,numbered 21-1,and the yield was 7.42 mg/L.The single factor experiment of inorganic salts and the simple optimization compared with orthogonal experiment were carried out in this strain,the yield of this strain was 8.55 mg/L,and the strain of the first sieve was increased by 15%.The characteristic absorption peaks of carotenoids produced by these 7 strains were compared with those ofβ-carotene,it was found that one of the contents wasβ-carotene.
