摘要
目的:研究肝经郁热型干眼患者与健康志愿者泪液之间的蛋白质组学差异。方法:纳入肝经郁热型干眼患者与健康志愿者各5例,采用数据非依赖性采集高分辨率色谱-质谱技术(DIA LC-MS)对肝经郁热型干眼患者与健康人泪液进行检测,筛选出差异蛋白,并对差异蛋白进行GO富集、KEGG通路、蛋白互作用网络等生物信息学分析。结果:干眼组与健康组比较共127个差异蛋白,其中115个上调,12个下调。肝经郁热型干眼患者差异蛋白主要涉及补体激活、经典途径,吞噬作用中的Fcγ受体信号通路等生物过程;涉及细胞外泌体、细胞外区域等细胞组分;涉及抗原结合、免疫球蛋白受体结合等分子功能。差异蛋白主要涉及PI3K-Akt信号通路、Fcγ受体介导的吞噬作用、原发性免疫缺陷等。肝经郁热组的重要节点蛋白有表皮生长因子受体、热休克70 kDa蛋白4、钙网蛋白等。结论:肝经郁热型干眼患者与健康志愿者泪液之间存在生物学差异,ENPP1有望成为肝经郁热型干眼的泪液生物标志物。
Objective:To study the proteomic differences between tears of dry eye patients with liver meridian stagnation heat and healthy volunteers.Methods:Five dry eye patients with liver meridian stagnation heat and five healthy volunteers were enrolled.The tears were detected by data independent acquisition liquid chromatography-mass spectrometry(DIA LC-MS),and the differential proteins were screened out.The bioinformatics analysis of GO enrichment,KEGG pathway and protein interaction network were carried out.Results:Compared with the healthy group,there were 127 differential proteins in liver meridian stagnation heat group,of which 115 were up-regulated and 12 were down-regulated.Differential proteins in dry eyes patients with liver meridian stagnation heat mainly involved in biological processes such as complement activation,classical pathway,Fcγreceptor signaling pathway;Involved in cellular components such as extracellular exosome,extracellular region;Involved in molecular functions such as antigen binding,immunoglobulin receptor binding.The main differences of proteins involved in PI3K-Akt signaling pathway,Fcγr-mediated phagocytosis,primary immunodeficiency and so on.Important node proteins in liver meridian stagnation heat group include epidermal growth factor receptor,heat shock 70 kDa protein 4,calreticulin and so on.Conclusion:There are biological differences between dry eye patients with liver meridian stagnation heat and healthy people.ENPP1 is expected to become a tear biomarker for dry eye with liver meridian stagnation heat.
