清肺达原颗粒治疗肺纤维化早期大鼠的作用机制 被引量:1

Mechanism of Qingfei Dayuan Granules in the treatment of early-stage pulmonary fibrosis in rats

摘要 目的:观察清肺达原颗粒对大鼠肺纤维化早期的干预作用并探究其作用机制。方法:30只SPF级雄性Wistar大鼠随机分为正常组、模型组和早期干预组,每组10只。气管注射博来霉素复制肺纤维化大鼠模型,术后第5天,早期干预组大鼠灌服清肺达原颗粒溶剂(32.55 g/kg),正常组和模型组大鼠灌服等体积0.9%氯化钠溶液,连续5d。1%戊巴比妥钠腹腔注射麻醉后取材。HE和Masson染色观察肺组织病理形态,ELISA法检测肺组织白介素-6(IL-6)、E-钙黏蛋白(E-cad)和α-血管平滑肌肌动蛋白(α-SMA)表达,Western blot法检测肺组织TGF-β1、p-Smad2、Smad2及Smad7蛋白表达。结果:与正常组比较,模型组大鼠肺组织IL-6、α-SMA含量及TGF-β1、p-Smad2蛋白表达水平均显著升高(P<0.01),E-cad含量和Smad7蛋白表达水平均显著降低(P<0.01);与模型组比较,早期干预组大鼠肺组织IL-6、α-SMA含量,以及TGF-β1、p-Smad2蛋白表达水平均显著降低(P<0.05,P<0.01),E-cad含量和Smad7蛋白表达水平均显著升高(P<0.01)。结论:清肺达原颗粒的早期干预能够减轻肺纤维化模型大鼠的肺组织炎症反应,减轻肺纤维化的程度,其机制与抑制TGF-β1/Smad信号通路的过度活化相关。 Objective:To observe the early intervention effect of Qingfei Dayuan Granules on pulmonary fibrosis in rats and probe its mechanism.Methods:Thirty SPF male Wistar rats were randomly divided into normal group,model group and early intervention group,10 rats in each group.The pulmonary fibrosis model was established by intrabronchial injection of bleomycin in rats.On the 5th day after surgery,rats in the early intervention group were given Qingfei Dayuan Granules decoction(32.55 g/kg)intragastrically,and rats in the normal group and model group were given equal volume of normal saline intragastrically,for 5 consecutive days.Rats were anesthesiaed by 1%pentobarbital sodium and sacrificed.HE and Masson staining were used to observe the pathological morphology of lung tissues.The expressions of IL-6,E-cad,andα-SMA in lung tissues were analyzed by ELISA.The protein expressions of TGF-β1,p-Smad2,Smad2 and Smad7 were detected by Western blot.Results:Compared with the normal group,the contents of IL-6 andα-SMA as well as the expression levels of TGF-β1 and p-Smad2 protein in the model group were significantly increased(P<0.01),while the E-cad content and Smad7 expression were significantly decreased(P<0.01).Compared with the model group,the IL-6 andα-SMA content as well as TGF-β1 and p-Smad2 expressions in the intervention group were significantly decreased(P<0.05,P<0.01),while the E-cad content and Smad7 expression were obviously ascended(P<0.01).Conclusion:The early intervention of Qingfei Dayuan Granules can reduce the inflammatory response and the degree of pulmonary fibrosis in pulmonary fibrosis model rats,its mechanism is related to the inhibition of TGF-β1/Smad signaling pathway.
