西双版纳野生猴群芽囊原虫感染的分子调查 被引量:2

Molecular investigation of Blastocystis infection in wild monkeys in Xishuangbanna

摘要 为了解西双版纳州原始森林公园野生猴群芽囊原虫的感染情况及其基因型,对采集的30份猴粪样,利用套式PCR法扩增芽囊原虫核糖体小亚基RNA(SSU rRNA)基因,对阳性产物进行测序比对,利用MEGA 6.0软件构建遗传系统进化树。结果显示:芽囊原虫检出率为40.00%(12/30);扩增的SSU rRNA基因序列长度为1100 bp,基因亚型鉴定为ST1和ST8,有4份样品比对结果显示为人芽囊原虫,其序列与人芽囊原虫一致性达99.61%。研究结果提示,加强对西双版纳原始森林公园野生猴群芽囊原虫的防控,确保猴群健康,具有重要公共卫生意义。 To identify the infection status and the genotypes of Blastocystis that infected wild monkeys of the Pristine Forest Park in Xishuangbanna,30 faecal samples were collected to amplify the small subunit ribosomal RNA(SSU rRNA)gene of Blastocystis by nested PCR.The positive amplified products were sequenced and the phylogenetic tree was constructed by MEGA 6.0.The results showed that the detection rate of Blastocystis was 40.00%(12/30).The amplified SSU rRNA genes were all 1100 bp in length,and the gene subtypes were identified as ST1 and ST8.The comparison of the four samples showed that the amplified sequences of Blastocystis were 99.61%consistent with those of the Blastocystis hominis.The present results indicated that there was great public health significance in strengthening the prevention and control of Blastocystis infection in the wild monkeys in the pristine forest park of Xishuangbanna so as to ensure the health of the monkey population in the area.
