摘要
为研究离蕊金花茶花粉活力、适宜培养基及花粉贮藏最佳条件。本试验以离蕊金花茶花粉为材料,采用3种方法进行花粉活力测定,用不同浓度硼酸和蔗糖的培养基进行花粉萌发培养,研究了花粉贮藏的适宜温度。结果表明:(1)I_(2)-KI染色法和TTC染色法测定结果均大于花粉萌发法,两者均适合测定离蕊金花茶花粉的萌发;(2)离蕊金花茶花粉的最佳培养基配方为1%琼脂+100 mg/L硼酸+200 g/L蔗糖;(3)离蕊金花茶花粉在不同贮藏温度下活力随时间的延长呈逐渐下降趋势,常温条件下花粉活力下降较快,而-20℃贮藏条件下活力保存最好,保存时间在1个月左右。
To study the viability,suitable medium and optimal storage conditions of pollen of Camellia liberofilamenta,the pollen viability of Camellia liberofilamenta was determined by three methods,the pollen germination was cultured in the medium of different concentrations of boric acid and sucrose,and the suitable temperature for pollen storage was studied.The results showed that:(1)the viability determination results of I_(2)-KI staining and TTC oxidation reduction method were better than that of germination method,and both of them were suitable for the determination of pollen germination vigor;(2)The best medium for C.liberofilamenta pollen was 1%agar+100 mg/L boric acid+200 g/L sucrose;(3)The pollen viability of C.liberofilamenta decreased gradually with the extension of time at different storage temperatures.The pollen viability decreased rapidly at room temperature,while the viability was best when preserved at-20℃for about one month.
