ICP22BP Affects the Proliferation of Glioblastoma Cells by Regulating Wnt Signaling Pathway

摘要 目的 分析胶质母细胞瘤(GBM)中相关调控基因,为其临床治疗提供一定的理论依据。方法 收集了86例GBM患者为研究对象,取全部患者的肿瘤组织及癌旁组织;并将A172细胞进行培养和转染,48 h后利用荧光定量PCR和蛋白质印迹法检测基因表达情况,通过CCK8实验检测细胞增殖情况。结果 GBM肿瘤组织中的ICP22BP mRNA及蛋白表达水平要显著高于癌旁组织,差异具有统计学意义(P<0.05)。ICP22BP表达减少会显著抑制A172细胞的增殖;A172细胞中敲低ICP22BP的表达可以显著抑制wnt信号通路下游相关基因(cMYC,c-JUN,WISP1和PPARD)表达。结论 胶质母细胞瘤癌组织的ICP22BP表达量要显著高于癌旁组织中的表达量。且ICP22BP下调后可以抑制Wnt相关信号通路下游基因的表达并抑制A172的细胞增殖。 Objective To analyze the related regulatory genes in glioblastoma(GBM) in order to provide a theoretical basis for its clinical treatment.Methods 86 patients with GBM were collected.The tumor tissues and adjacent tissues of all patients were removed;A172 cells were cultured and transfected.After 48 hours,the gene expression was detected by fluorescence quantitative PCR and Western blot,and the cell proliferation was detected by CCK8 experiment.Results The expression levels of ICP22 BP mRNA and protein in GBM tumor tissues were significantly higher than those in adjacent tissues(P<0.05).The decreased expression of ICP22 BP significantly inhibited the proliferation of A172 cells;Knockdown of ICP22 BP expression in A172 cells can significantly inhibit the expression of related genes downstream of Wnt signaling pathway(cMYC,c-JUN,WISP1 and PPARD).Conclusion The expression of ICP22 BP in cancer tissues was significantly higher than that in adjacent tissues in glioblastoma patients.The down-regulation of ICP22 BP can inhibit the expression of genes downstream of Wnt related signal pathway and inhibit the proliferation of A172 cells.
