壳寡糖复合固体饮料对高尿酸血症小鼠肠道微生态的调节作用研究 被引量:3

摘要 为探究壳寡糖复合固体饮料(KSP)对高尿酸血症小鼠肠道微生态和短链脂肪酸的影响。将小鼠分为正常对照组、模型组、低/中/高剂量KSP组、别嘌呤醇组,通过灌胃酵母膏和腹腔注射氧嗪酸钾建立高尿酸血症模型,通过喂食含KSP的饲料给药。21 d后收集小鼠血清检测尿酸,收集结肠、盲肠内容物用于短链脂肪酸检测和16S rDNA高通量测序。结果显示:KSP可以降尿酸和改善拟杆菌属(Bacteroides)、拟普雷沃菌属(Alloprevotella)、普雷沃氏菌科UCG-001(Prevotellaceae_UCG-001)、阿克曼氏菌属(Akkermansia)、乳酸菌属(Lactobacillus)等的相对丰度,同时缓解由高尿酸血症引起的丙酸、丁酸、戊酸、异戊酸等水平的异常增高。因此该壳寡糖复合固体饮料可以通过调节肠道菌群组成和短链脂肪酸水平改善高尿酸血症。 The present study explored the effects of chitooligosaccharide compound solid beverage(KSP)on intestinal microecology and the production of short-chain fatty acids(SCFA)in hyperuricemia mice.Mice were divided into a normal control group,a model group,low-,medium-,high-dose KSP groups and an allopurinol group.The hyperuricemia model was induced by intraperitoneal injection of potassium oxonate and intragastric administration of yeast extracts.The mice in the KSP groups were fed with fodder containing KSP.After 21 days,serum samples were collected to test the level of uric acid.The colonic and cecal contents were collected for SCFA detection and 16S rDNA high-throughput sequencing.The results showed that KSP could lower the uric acid level and regulate the relative abundance of Bacteroides,Alloprevotella,Prevotellaceae_UCG-001,Lactobacillus and Akkermansia in hyperuricemia mice.Additionally,KSP could also improve the abnormal increase in propionic acid,butyric acid,valeric acid and isovaleric acid induced by hyperuricemia.The findings of this study revealed that this chitooligosaccharide compound solid beverage could improve hyperuricemia by adjusting the composition of the intestinal flora and SCFA level.
